| In this paper, the anti-tumor mechanism of isoflavones was introduced briefly, Phenoxodiol, the anti-tumor candidate, which was the derivative of isoflavone. was introduced in detail. Its preclinical study and the progress of clinical research were described in this paper. The synthetic process of Phenoxodiol was studied in this paper. Bases on four synthetic routes of Phenoxodiol reported in literatures and after experimental testing, the synthetic route of Phenoxodiol was designed as below:the target compound was prepared via transfer hydrogenation, protection, reduction, dehydration, deprotection with Daidzein as the starting material. Four different protecting groups of 4’,7-hydroxy at isoflavone nuclei, such as acetyl, MOM, methyl or benzyl were introduced. The results showed MOM group was the preferred one.The structure of the above obtained target product was confirmed by1H-NMR,13C-NMR and MS. Its melting point was consistent with that reported in literature. The total yield was 46.8%. The HPLC purity was above 99%. |
