Synthesis And Immunological Activity Of Fully Synthetic Two-component Anti-tumor Vaccines With αGalCer As An Adjuvant

Among the new therapies against cancer,immunotherapy remains one of the most promising therapies.With the development of immunotherapy,by using it in combination with chemotherapy/radiation therapy or alone,it can effectively inhibit tumor growth and even kill tumor cells.Compared to normal cells,tumor cell surfaces often express abnormal glycosylation,which provides a suitable target for the development of anti-tumor carbohydrate vaccines for cancer immunotherapy.However,the main problem is the weak immunogenicity of tumor-associated carbohydrate antigens.There are two parts about this article.The first part was to synthesize a simple two-component cancer vaccine,the tumor-associated Tn saccharide antigen was covalently conjugated with iNKT cell agonist.Immunological evaluation of this vaccine based on Tn-Thr-aGalcer or Tn-Thr-pGalcer was explored after immunization with mice,the effectiveness and feasibility of this strategy were discussed later;The second part was demonstrated that a structure-modified Tn antigen analogue was chemically synthesized,the modified Tn antigen was conjugated to the iNKT cell agonist for immunization studies,the effects of Tn mimetic on enhancing the immunogenicity of the Tn antigen was investigated.Immunological results of these vaccine indicated that Tn glycoforms can be recognized by the immune system,and both types of vaccines can elicit high titers of antibodies,drive cytokine release,and obtain efficient immunization.However,the cross-recognition ability of antibodies triggered by Tn mimetic needs to be further improved.
