Red kermel color is an important natural edible pigment. It has the characteristics of edibleness, innocuousness, free of side effects and high nutritional value. Recently, a large number of researches have shown that red kermel color had a lot of biological activities such as antioxidant, lowering blood pressure, anticancer, antianaphylaxis, etc. This paper is to evaluate the anti-aging effect of red kermel color in vivo, and to provide a theoretical basis for the future popularization and application of red kermel color.Firstly, using cyanidin-3-glucoside as a standard, red kermel color anthocyanin which is the main functional ingredient had been quantitated by HPLC-ESI-MS. The column was Thermo Hypersil GOLD C18 column (100×2.1 mm). A mixture of acetonitrile and H2O-1% formic acid solution was used for elution. Three anthocyanins were identified. They were cyanidin -3,5-diglucoside 0.99 mg/mL, cyanidin-3-glucoside 16.77 mg/mL and peonidin-3-glucoside 1.41 mg/mL, respectively.Secondly, the antioxidant ability of red kermel color in vitro was studied. Red kermel color showed stronger ·OH scavenging ability and anti-lipid peroxidation. Besides that, it had some of reducing power, · DPPH scavenging ability and anti-erythrocyte hemolysis ability, but the abilities were weaker than vitamin C.Thirdly, to further investigate the antioxidation and anti-aging ability of red kermel color, we selected D-galactose aging mice. Aging model mice were established by cervicodorsal region subcutaneous injection with D-galactose continuously. The results showed that red keemel color could significantly reduce methane dicarboxylic aldehyde (MDA) content in serum, brain, liver, kidney of aging mice induced by D-galactose and increase super oxygen dehydrogenises (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) activities in serum, brain, liver and kidney, only GSH-Px activity in brain was not significant, the rest were significant differently. Monoamine oxidase (MAO) activity of red kermel color of three dose-groups decreased in the three organs. Through the observation of mice brain tissue pathological, red kermel color improved neuronal cells form and number state.Finally, the experiment carried through the preliminary discussion to anti-aging mechanism of red kermel color, by real-time PCR determination of the expression levels of mouse liver SOD I, SOD2, MAO-B mRNA and related enzyme activities.The results of experiments showed that red kermel color possess strong antioxidant and anti-aging activity.
