This study use the Drosophila melanogaster as experimental animal model to evaluate the antioxidant and anti-aging function of lutein, so that our study can provide theoretical basis for the further research and application of lutein.We used the HPLC method to detect the purity and content of the lutein sample. According to the standard curve and results of sample peak area, the content of lutein is 85.99%.We studied the the effect of lutein on anti-aging and gene expressions of antioxidant enzymes of drosophila in normal group. The results show that the flies treated by lutein with the longer lifespan compared with that of the control group. With the doses of lutein increased in the experiment, the activities of antioxidant enzymes were increased while the MDA content was reduced. The gene expressions of antioxidant enzymes in drosophila were analyzed when the flies were reared with lutein at 25 d. Results showed that the mRNA gene expressions of CuZn-SOD, Mn-SOD and CAT of drosophila fed with lutein were significantly increased compared with that of control group (P<0.05). Thus, the research indicates lutein has the obvious capacity of antioxidant.Besides, the anti-aging effect of lutein in drosophila damaged by UV and D-galactose were studied. The results of survival test and biochemical test of oxidative damage drosophila fed with different doses of lutein showed that, lutein has the capacity of prolong the lifespan of flies and increased the activities of CnZn-SOD, Mn-SOD and CAT, and reduced the content of MDA.At last, the anti-aging effect of lutein in drosophila damaged by H2O2 and Paraquat also be studied. The two acute test results show that the lutein could extend the lifespan of drosophila compared with the flies fed with basal diet, and the results present a visible relationship between dose and response.The results show that lutein has good antioxidant activity ability, the lifespan-prolonging activity of lutein was partially by up-regulation of endogenous antioxidant enzymes.
