Water chestnut is a widely cultivated aquatic free-floating plant in China. Water chestnut is popular Chiese traditional edible vegetable. As processing by-products, the pericarps are commonly considered to be waste and polluted to environment. Recent studies showed that pericarps of many fruits and vegetables contain numerous bioactive compounds, such as polyphenols, flavonoids and alkanoids, etc. Flavonoids is a variety of health care of material such as tumor suppressor and antibacterial. In order to take advantage of water chestnut shell resource, in this paper, the pericarps of water chestnut shell from Hubei was analyzed extraction technics, purification method and activity evaluation. Mainly results were list as follows:1. Optimization of extraction process of flavonoids from water shell. The application of traditional extraction flavonoids extraction water chestnut shell. Using single factor experiment and Box-Benhnken center, The optimized water chestnut shell optimum parameters of flavonoid extraction. The optimum extraction conditions were 60 ℃, 30 min,50% and 1:30(mL/g) for temperature, ultrasonic treatment time, ethanol concentration and liquid-to-solid ratio. Under these conditions, the actual and predicted yields of flavonoids from water chestnut hull were 2.99% and 3.08%, respective, 3.01% higher than theory predicts.2. Study on purification method of flavonoids from water caltrop shell. The results showed that resin AB-8 is the best for the purification of flavones. Furthermore, the volume of sample is 110 mL, the flow rate is 0.8mL/min and the eluent is 70% ethanol, the velocity of0.8 mL/min, the volume of 110 mL, 340 mL70% ethanol elution agent solution, the elution flow rate 0.8 mL/min.Water chestnut shell flavonoids crude extract purity of 17.5% and purification purity of 36.75%.3. Water chestnut shell flavonoids extracts of bacteriostatic activity of study. Flavonoids from water chestnut hull have remarkable antimicrobial effected on pseudomonas and staphylococcus. Water chestnut shell the crude flavonoids extracts extractings bacteriostatic MIC value is 3.125、12.0mg/m L, pured flavonoids extracts extractings bacteriostatic MIC value is 3.125mg/mL. Based on the liquid water chestnut hull growth curve analysis, the results showed water chestnut shell the crude flavonoids extracts had stonger inhibitory effect.The grow of Pesudomonas on the duck meat medium under different temperature increased the LPD can reflect the Pseudomonas significantly increased, conclusion: Kojc acid inhibition effect of better than water chestnut shell flavonodis purification.4. Water chestnut shell flavonoid extracs research on inhibition of tumour cells. Waterchestnut shell coarse extract, purification of flavonoids can inhibit human gastric cancer cells SGC7901 and human liver cancer cell proliferation of Hep G2. The results showed that water chestnut shell the crude flavonoids extracts and pured flavonoids extracts have an effect on human gastric cancer cells and Human liver cancer cells Hep G2, especially liver cancer cells Hep G2, IC50 alue concentration of 124.9ug/mL,R2=0.95.
