Abstract/Summary:
Pumpkin peel(PP) known as processed disposal of pumpkin which been studied for Anticancer, Antioxidant and among other functions. The main research about the active constituents of PP were take place on determination of total polysaccharide, pectin etc. Although that PP pigment related research is still in the preliminary stage for now. And that gives us privilege to run a systematic analysis for PP’s phytochemicals, pigment extraction process and its stability, Anti-oxidant activity, purification process in vitro. The results were as follows:pp contains:protein, amino acids, peptides, carbohydrates, saponins etc; anthraquinone, cardiac glycoside, fats has not been detected.Use the traditional organic solvent refluxing extraction method extract the pigment of pumpkin peel, the optimum condition obtained for the response surface method is: extraction time was 1h, solid-liquid ratio was 1:20, extraction temperature was 60℃, 90%ethanol as extraction agent, extraction one times.PP pigment has a poor light stability; Can’t be in long time exposure, during storage should avoid direct sunlight; In the maximum security scope the common food additives as citric acid, sucrose, glucose, soluble starch is almost no effect on the stability, can be used as the colorant. Stable for the metal ions of Mg2+, Na+, Ca2+, according to the condition its apply in practice; tolerant to antioxidant agent, stable for acid and alkali, but can not be apply extremely strong acid environment.PP pigment have the ability to remove the·OH, ·DPPH, O2-; and also total reducing power is comparatively well, but compared to the same concentration of ascorbic acid, the antioxidant activity lower than ascorbic acid, this may be attributed the pigment extracts has not be purified; Means that further research for purification of pp pigment have more practical significance.pp pigment crude extract components are so complex, contains many impurities; this experiment use the macroporous resin adsorption method, purifies the pp pigment, the optimum condition is: D101 macroporous resin as adsorbent agent, static adsorption time was 3h, herbs-resin ratio was 1:7, pH was 4, Ander this condition the maximum adsorption rate reaching 75.3%, static equilibrium time was 90 min, eluent agent was 60%ethanol, sample flow rate was 5mL/min, elution flow rate was 1mL/min.Finally, Use the TLC and HPLC method, the PP Pigment purified products were preliminary separated and quantitative analysis, as a result: pumpkin peel pigment contains lutein and beta carotene, lutein was 16.80μg/μL, beta carotene was 18.01...
