Polysaccharide is very important bioactive substances widely found in a variety of organisms,in recent years, with the deepening of the polysaccharide study, people in multi-carbohydrate research, development and utilization has also made great progress. Now polysaccharide widely used in medical, health care and so on. Pumpkin has a lower blood sugar, blood pressure,lipid-lowering effect was widespread awareness. But research on the pumpkin active polysaccharides is not deep enough, therefore, the subject of pumpkin polysaccharide first by extraction and purification process to start, optimization methods of extraction and purification of pumpkin polysaccharide isolated, on the basis of its preliminary structural identification the study.The main experimental results are as follows:1. by cellulase extraction, hot water extraction method, ultrasonic assisted extraction method pumpkin polysaccharide experimental extraction rate for the optimal solution for each test method pumpkin polysaccharide extracted for comparison, we can see extract cellulose extraction higher extraction rate pumpkin polysaccharides. I.e. an enzyme concentration of 0.5%, a time 120 min,p H 5.2 the extract, the extraction temperature was 55 ℃. The use of the optimal solution for extraction, the extraction rate of up to 4.13% pumpkin polysaccharides.2. by choosing AB-8 macroporous resin as pumpkin polysaccharide purification materials,respectively, of its static and dynamic adsorption experiments. Effects of various factors on the static and dynamic single adsorption results showed that: static adsorption, the purity of pumpkin polysaccharide was 19.73±3.52, while the dynamic adsorption by its purity of 33.58±1.03. It can be seen, with AB-8 adsorption material, the use of dynamic eluting pumpkin polysaccharide purification.3. the use of pumpkin polysaccharide gelcolumn chromatography separation, by UV and IR chromatography after extraction and purification of polysaccharides were identified, confirmed the higher pumpkin polysaccharide extract purified purity, the physical constants, chemical and spectral analysis and identification of pumpkin polysaccharide sugar chains by furanose and pyranose components.
