Biotin, also known as vitamin B7, vitamin H, Coenzyme R, is one of water-solubility vitamin. Since it can not be synthesized by human beings, it is regard as an essential nutrient for human body. Biotin can be obtained by diet or be synthesized by gut microbes. However, the intestinal microbial ecosystem of infants still not be well established, the biotin can only be supplemented by diet.Chromatography and microbial method are the two mainly method for biotin determination.Chromatography is a more accurate, stable and lower detection limit method than that of microbial method. In contrast, microbial method is higher selectively, more convenient and lower cost.In this paper, the HPLC and Lactobacillus plantarum methods were employed to determination the biotin content in infant foods. To improve the detection sensitivity, the conditions, the reagent, steps, parameters, et al, for detection were optimized and the biotin contents of 4 infant foods were determined. The advantages and disadvantages of two methods were compared based on regression curve R2, determination results, RSD, recovery rate, and even cost, time consumption, the operation.According our data, we found that methanol was not the good regent for biotin detection and we selected the mobile phage buffer as acetonitrile-0.050 mol/L KH2PO4(water: acetonitrile15:85, p H 3.4). the results showed that R2= 0.9997, the average RSD was 2.86 and the top RSD was 3.91 for HPLC method. On other hand, the R2 was 0.9958, the average RSD was 2.61 and the top RSD was 2.87 for the Lactobacillus plantarum method under optimization condition.The result demonstrated that two biotin detected method has a linear relationship and good stability, the detection limit is low enough. Despite two methods have their own advantages and disadvantages, they are both suitable for determination the biotin content in infant food.
