1. In this paper, thirty nine new target compounds were designed and synthesized, including thirty mono-1,2,4-triazine derivatives I-56 and 9 bis-1,2,4-triazine derivatives II-45. The structures of the target compounds were characterized by IR, 1H NMR, elemental analyses and single crystal X-ray diffraction.2. The optimum experimental conditions of preparing mono-1,2,4-triazine derivatives I-56 under the microwave irradiation were found. This synthetic method has such advantages as short reaction time, simple operation, higher yield comparing with classical methods.3. The synthesized thirty target compounds I-56 were evaluated for their inhibitory activities against Cdc25 B and PTP1 B. The results show that most of them display remarkable inhibitory activities against Cdc25 B and PTP1 B. Among them, compound I-6o exhibits highest inhibitory activity against Cdc25 B [IC50=(0.84 ± 0.22) μg/m L]. Compounds I-5i, I-5m, I-5n, I-6h, I-6j, I-6m, I-6n and I-6o exhibit more potent inhibitory activities against PTP1 B [IC50=(0.46 ± 0.10)0.87 ± 0.19 μg/m L] than reference compound oleanolic acid [IC50=(0.97 ± 0.15) μg/m L]. It is noteworthy that compounds I-5h, I-5m, I-6n and I-6o exhibit higher inhibitory activities against both Cdc25 B and PTP1 B. They can be considered as potential Cdc25 B and PTP1 B inhibitors, and have great application prospects in the treatment of cancers and diabetes.4. The recognition properties of target compounds II-4b and II-5b with metal ions were investigated by UV-vis and fluorescence spectra. The results show that Cu2+ and Fe3+ ions display the fluorescence quenching to compound II-4b. However, Sn2+ ion displays the fluorescence enhancement to compound II-4b. Thus, compound II-4b exhibits the selective recognition to Cu2+, Fe3+ and Sn2+ ions. Cu2+, Fe2+ and Fe3+ ions display the fluorescence quenching to compound II-5b. Therefore, compound II-5b exhibits the selective recognitions to Cu2+, Fe2+ and Fe3+ ions. Target compounds II-4b and II-5b have potential application in the detection of Cu2+, Fe2+, Fe3+ and Sn2+ cations.5. The results in this paper provide a scientific basis for the design, synthesis, structural characterization and drug development of the new anticancer and anti-diabetic drugs, and the development of cation recognition receptors. Therefore, this study has great significance.
