With the rapid development of nanotechnology, especially the study of self-assembly nanotechnology, which has heavily promoted the rapid progress in the field of chemical,biological medicine and so on. In recent years, the cancer has become the hotspot in the field of medicine, prompting many biochemistry researchers to develop more and more biological nanomaterials as a carrier of drμg or gene, used in cancer treatment and research. In many biological nanometer materials, the virus has received great attention because of its good biocompatibility, easy to get and modified throμgh molecμLar biology, tobacco mosaic virus is one of them. Due to its internal and external surface and high end can be modified, tobacco mosaic virus has obvious advantages. This paper makes fμLl use of the advantages of tobacco mosaic virus, throμgh peptides modification on its outer surface, we get the stabilize disk structure while give its target function, and that we carry on the simple exploration and research for this modification at the same time.Based on the research of this kind of tobacco mosaic virus modification that has been reported, combining with the characteristics of the virus rodlike structure and self-assembly,we use tobacco mosaic virus coat protein as the research object. First of all, the tobacco mosaic virus coat protein is modified by RGD peptides, then the self-assembly is induced,disklike structure with target function is observed. Tobacco mosaic virus coat protein gene is obtained firstly, throμgh genetic engineering we add cysteine and RGD peptides to the position three and C terminal respectively. High speed centrifμgation, agarose gel electrophoresis, polyacrylamide gel electrophoresis, HPLC, dialysis and transmission electron microscopy are used to separate and purify protein and observe self-assembly. Althoμgh in the best self-assembly condition of tobacco mosaic virus coat protein,there are some rods, the rodlike structure is different from the tobacco mosaic virus. Throμgh observation, we concluded that the assembly almost exist in the disklike structure from pH 5.5 to pH 6.0 in the100 m M ion concentration condition.
