| Natural products have become a source of inspiration for pesticide lead compounds due to the rich structure,specificity to target species,unique mode of action,and biodegradability.In recent years,scientists have found that natural product ferulic acid has good antiviral activity,but its inhibitory effect on plant viruses is not ideal.Therefore,in order to develop high-efficiency,low-toxicity and environment-friendly small molecule anti-plant virus drugs,a series of ferulic acid derivatives containing acyl(sulfo)amide active units were designed and synthesized based on ferulic acid by using the principle of active splicing.At the same time,cucumber mosaic virus(CMV)and tobacco mosaic virus(TMV)were used as research objects to test the biological effects of compounds C1-C6,D1-D7 and E1-E21 on the two plant virus diseases.Based on the good curative activity of compound E2,the antiviral mechanism of compound E2was studied by the determination of chlorophyll content,transcriptomics and proteomics.The main research work are as follows:1、A series of novel ferulic acid derivatives containing acyl(sulfo)amide units were designed and synthesized through the principle of active splicing based on the ferulic acid backbone,and their structures were identified by 1H NMR,13C NMR and HRMS.2、The aniviral activities in vivo of target compounds to CMV and TMV were tested by the half leaf spot method.The results of biological activity test showed that some ferulic acid derivatives showed good anti-plant virus activity against CMV and TMV.Among them,compound E2(401.7 and 189.0μg/m L),E4(446.9 and 260.2μg/m L),E8(429.3 and 299.1μg/m L)and E16(472.7 and 266.3μg/m L)showed good curative activity against CMV and TMV.In addition,compound E2(431.2,291.8μg/mL and 187.0,169.2μg/mL)showed outstanding protective and inactivating activities to CMV and TMV,which were superior to the positive control agent ribavirin(652.6,427.4μg/m L and 589.7,359.4μg/m L)and ningnamycin(457.1,318.7μg/m L and 268.1,237.6μg/m L).3、To explored the antiviral mechanism of compound E2,the changes of chlorophyll content,differentially expressed genes and proteins in tobacco treated with compound E2 were analyzed.The experimental results showed that compound E2 can increase the content of chlorophyll in plants and promote plant photosynthesis.Meanwhile,compound E2 can obviously regulate the expression of genes and proteins,and mainly controls the photosynthesis of the plants,thereby improving the disease resistance of the plants. |
