It’s our first time to find the model plant species Arabidopsis thaliana in Qinghai-Tibet Plateau which is called "the world’s third pole"Firstly, this study discussed about the identification and classification methods on the basis of founding the Arabidopsis thaliana ecotype. According to the available methods, living conditions, altitude, phylogenetic tree and the result of this study, We dare to raise an idea that, the Arabidopsis in Qinghai-Tibet Plateau is likely to be a new Arabidopsis thaliana ecotype which is never discovered before.Secondly, on the basis of Arabidopsis genome sequencing Tibet individual, we took Col0 as a reference and extracted sequence of Arabidopsis 131 SINEs from Tibet individuals. Then we conducted a statistic of each presence and absence of a SINE from 20 kinds of Arabidopsis thaliana ecotype in the known genome sequences. We screened Arabidopsis ecotype at least one missing in 40 SINEs. And on this basis, these 20 kinds of Arabidopsis thaliana ecotype can be distinguished by the selecting combination of the two groups SINEs.Finally, based on the whole-genome sequencing, as for the ambiguities 30 SINEs in the whole genome sequencing, primers were designed around 100-600 bp within its scope using Can0 or Bur0 SINEs in full genome sequence as a template, the amplification of the corresponding fragments and were sent for sequencing. Compared with the whole genome sequencing, the obtained sequencing can show a further validation that preparatory and obtain sequence information have not read the whole genome sequencing.
