Rhythm is one of the most basic characteristics of biology. Earth rotation produce alternating day and night and in the process of constantly adapt, Itself also generates a cyclical changes. From bacteria to higher mammals, biological differentiate into special timing system in a long adaptive evolution.The biological clock control activities and behavior and periodic changes.Myospalax cansus life in the darkness of the underground.There is degeneration of the visual system in long-term adaptive evolution and no function of visual imaging, but can identify the light and dark environment. Molecular biology of circadian rhythm is a series of clock gene transcription and translation activities and It form the oscillation of the feedback loop. Clock as a positive adjustment component and play a role in the molecular oscillation mechanism. Through ENU get circadian rhythm mutant mice,CLOCK protein abnormal expression and circadian rhythm in mice, including rhythm disappeared and endogenous rhythms change, and prove the importance of Clock genes in mammalian circadian rhythms.In Myospalax cansus degradation of the visual system, Clock play a role for the regulation of circadian rhythm.Clock mRNA expression and regulation circadian rhythm has not be in-depth study. This experiment using tissue section to observe the microstructure of harderian gland. Real-time PCR technology to test harderian gland and other tissues Clock mRNA expression in DD and LD and to discuss Clock regulating circadian rhythm. In order to research to the circadian rhythm of regulating mechanism in rodents and provide the reference. The main experimental results are:1.Observed the whole morphology of harderian gland in Myospalax cansus Harderian gland is very fat and surface of capsule.Surrounding the degradation of the eye and below the backward outspread (PlateⅠ:Fig.2).Harderian gland are located in the temporal and subtemporal fossae, under the covering of the zygomatic arch,and with temporal muscle,masseter muscle,dermal muscle(PlateI:Fig.2T M D). The Harderian gland resembles a creamy body (PlateⅠ:Fig.3)and about one over one thousand of the weight. Harderian gland and and its ratio with body weight is not significant difference in male and female.2. Using histological techniques to study the microstructure of harderian glands.The results show that harderian gland is tubuloalveolar with a clear division into lobules (PlateⅡ:Fig.1). The acini are composed of cuboidal cells and merge to form a wide secretion of pipeline (Platell:Fig.1C). The activity of the acini is usually equally divided among lobules, but in some cases there is a clear morphological division between2cellactivity forms. The thicker form is more common than the thin (PlateⅡ: Fig.4) and the relatively rare appearance of the thinner cells (PlateⅡ:Fig.2).The secretory cells have numerous lipid vacuoles of various sizes (PlateⅡ:Fig.3).3. Using real-time PCR technology detection Clock mRNA expression in Myospalax cansus harderian gland, eyes, liver and kidney.The results show that Clock mRNA expression level has no oscillation in0:00,6:00,12:00,18:00and circadian rhythm disappeared (P>0.05). The experimental results are consistent in DD and LD.The Clock mRNA expression level of different in different organizations.Harderian gland Clock mRNA expression is significantly higher than eyes,liver and kidney (P<0.05).Four tissue of Clock mRNA expression level in DD and LD has a significant difference (P<0.05).The above results show that Myospalax cansus eyes extreme degradation and still can feel light stimulation. Harderian gland can be used as a light receptor and accept light stimulation and participate in regulation of circadian rhythm, keep synchronous with the environment and adapt to underground life. In a word Myospalax cansus can accept the outside of optical signals.Through the retina signaling pathways to start the Clock transcription and translation activity and feel the light cycle.
