In order to realize the function of the observation and quantitative analysis of living cells for a long time and provide accurate and reliable experimental data for the cells kinematics, this thesis introduced the target tracking technology which was based on digital images processing to the field of living cells tracking.The images sequence of living Colon cancer cell lines that was captured by a system which mainly consist of an optical microscopy and an image sensor. This thesis has summarized and confirmed the main process of cells tracking. The study has been conduct on segmentation algorithms. A segmentation method with two steps was adopted, the thresholding to recognize the cells regions and the watershed segmentation algorithm which based on morphology to separate the adhesive cells. There were two kinds of algorithms were used to cells tracking in this paper, they were the mean shift tracking algorithm and the tracking algorithm based on topological constraint. The function of living Colon cancer cells tracking has been realized through improving these two algorithms. The cells’ trajectories and tracking errors were gotten through analyzing the experimental data. The experimental results have proved the applicability of two algorithms in the living cells tracking. At the same time, this thesis has summarized the advantages and disadvantages of the algorithms in the field of living cells tracking, confirmed the applications of the two algorithms respectively.
