Digital holographic microscopy technology combine digital holographic technologywith optical microscopy holography,CCD is used to record information for digitalholographic microscopy instead of the traditional holographic dry plate. It is simple andfast to record and digitalize the image,and using computer technology and digital imageprocessing technology can effectively improve the quality of reconstruction whenreconstruct the hologram.In nearly a decade of development, digital holographicmicroscopy has certain theoretical basis and some applications in medical,computerelectronics and fluid field,but some key technology needs further research in digitalmicroscopic holographic, the problem such as how to suppress or compensation the phasecurvature introduced by the microscope objectives, how to get the precise focal plane,how to improve the lateral resolution, how to get the object’s actual magnification, how todetermine the relationship between the reconstruction distance and the recordingdistance,how to realize the real-time stability measurement,these problems seriouslyrestrict the practical application of the technology.In this paper, we studied and improvedon some issues according to the above problem, it is great importance in the measurementof the microscopic objects in the microscopic world and fluid field.In order to study the relationship between the recording distance and the reconstructiondistance, this paper firstly researches the point light source sphere wave amplificationsystem.The principle of theoretical knowledge and the algorithm of the reconstruction ofthe point light source are researched.and analyses several experimental of the sphericalwave illuminated object and the advantages or disadvantages of several experiments,and chose the best experiments to analyses the relationship between the recordingdistance and the reconstruction distance. This chapter also analyses the some basicprinciples of microscope objectives imaging directly introduce in digital holographic,andfind some specific problems of the digital microscopic imaging and the reason of theseproblems by the combination of theory and experiment method.The distance between target and microobjective and the position of image through theobjective lens is difficult to accurately obtained in digital microscopic holographic.Themagnification of the hologram received by CCD is not the same as the magnification ofmicroscope but an uncertainty values.Based on the composition of lens as theory,the relationship between CCD and the image when the position of object is fixed and therelationship between the position of object and CCD when the position of CCD is fixed isdiscussed in this paper,magnification function equation and the relationship functionsamong the parameters and optimized experimental parameters in the optical path ofholographic microscopy is obtained in this paper under the premise that the image isclearly reconstructed.The experiment is Proposed and demonstrated,CCD can capturehigh-quality of the hologram and the hologram can reconstruct clearly when theconditions of function and optimization of the parameters is satisfied,the quality ofholograms and reconstruction is poor when the above relationship is notsatisfied.According to the above study,the adjustable magnification experimental systemis designed,the magnification and reconstruction distance are accurate value by adjustingthe distance between the lens,and experiments verify the accuracy of the magnificationand reconstruction distance.
