Microarray technology as a powerful biological analysis tool has been widely concerned and researched in recent years. Due to the expensive price of microarray spotting instrument and laser fluorescence scanner, the applications of microarray technology are currently restricted to well hospitals with advanced equipment and professional biological laboratories. As a consequence, it has not been widely popularized up to now. So it has an important signifitance to find a quick, low-cost and simple operation quantitative detection method, in public health and early disease diagnosis. With the development and popularization of the inkjet printer and optical drive/disk, The use of inkjet printing technology to fabricate microarrays on polycarbonate surface of DVD, combined with optical drive molecular detection technology, is expected to become a new generation of promising medical diagnostic tool. In this paper, we studied the preparation of digital versatile disc (DVD) biological disk based on inkjet printing method, we also detected the concentration of streptavidin based on optical drive signal identify method. It is worth mentioning that in the process of experiment, the devices are popular commercial products, so the advantage of this method is the detection of low cost, simple operation, easy promotion. In this paper, we mainly completed the following works:1. With the surface of plastic were successfully photo-activated by UV/Ozone cleaner, polycarbonate (PC) substrates of DVD were changed from hydrophobic to hydrophilic. The result showed that the contact angle of water on the PC was reduced from88.07°to9.33°.2. We prepared Polydimethylsiloxane (PDMS) micro-fluidic plates for confining the reaction area. Biotin and streptavidin were covalently bound to the disc surface in the channel. Signals were amplified through the use of a gold/silver staining treatment. DVD optical drive and disk diagnostic software are used for detecting errors, and it has the ability to quantify biochemical recognition reactions through analysing the PIF per unit area and optical darkness ratio. Comparing with the bio-CD, bio-DVD has an advantage in detection sensitivity.3. Commercial piezoelectric drop on demand (DOD) ink-jet printer without any modification was used to fabricate1μg/mL biotin multi stripe array on DVD specified section surface by controlling printer and ink diffusion function. The experimental results indicate that the smallest printing volume is500micro liters; the optimum printing times is five times; the detection sensitivity and dynamic ranges for streptavidin is0.6μg/mL0.6~1.2μg/mL.
