Objective To ascertain the optimal proportion of Z and H which have effect on myocardial hypertrophy, to demonstrate the rationality of the prescription, and to investigate its initial pharmacodynamics.Methods The myocardial hypertrophy model was established by administrating the ISO subcutaneously in mouse. The cardiac index was determined by using orthogonal-design method, to definite the optimal proportion of Z and H. In the pharmacodynamic experiment, the effect of the complex prescription which comprised the two ingredient in the optimal proportion, was investigated by measuring heart rate, running time, swimming time, survival time in hypoxia and cardiac index after the model was set up as above. Assays using the viability, activation of content of maleic dialdehyde(MDA) and superoxide dismutase(SOD) were chosen for measuring the degree of cardiac myocytes injury.Result The study reveals that Rh>Rz; the cardiac index was found to be the minimum in the ZH(25:100) treated group. It is observed that there was antagonistic role against increasing of the heart rate in Z.H.ZH treated group. The heart rate, running time, swimming time and survival time was found to be increased in the Z and ZH treated groups. It is also be revealed the antagonistic effect on myocardial hypertrophy pronouncedly in ZH treated group. Compared with the model group. ZH protected isoproternol-treated rat cardiac myocytes from death and significantly decreased MDA release and increased SOD activity.Conclusion ZH has the restrain effect on myocardial hypertrophy induced by Iso in mouse. Among the selected doses, the theoretical optimal proportion of A and B was 25:10. The complex prescription could enhance the action. ZH complex appears to have antixidation and protected effect from Iso-induced cell damage.
