ObjectiveTo explore the extracts of sea cucumber ovum influence of on the reproductive system of female mice and its possible mechanisms, offering theoretical basis for further development of celery.Methods1、The water extracts of sea cucumber ovum were made through ultrasonic auxiliary method, and the fat soluble material were extracted by ethanol.2、The contents of trace elements in lyophilized of sea cucumber ovum were detected by ICP-MS. Determination of moisture, ash, protein and fat content in lnyophilized of sea cucumber ovum according to GB5009standards.3、Ninety healthy female kunming mice were selected and randomly divided into9groups: control group, model group, low dose of water extracts group (150mg·kg-1), middle dose of water extracts group (300mg·kg-1), high dose of water extracts group (300mg·kg-1), low dose of ethanol extracts group (150mg·kg-1), middle dose of ethanol extracts group (300mg·kg-1), high dose of ethanol extracts group (300mg·kg-1), and the estradiol valerate group (0.085mg·kg-1). Except the control group injected with normal saline, the other were injected with28mg/kg cyclophosphamide daily for successive5days. Caused by endocrine disorders, premature ovarian failure occurs then. At the same time,the treatment group were given by different drugs once a day for4weeks. The general status, organ index, hormone levels in serum, histomorphology of ovary and uterus, and endometrial thickness were observed. Activities of SOD and GSH-Px in ovary and uterus were determined, contents of the total protein and MDA in ovary and uterus were determined, observe the apoptosis and expressions of apoptosis-associated genes with immunohistochemical method. Results1、The condition for the extraction of water extracts of sea cucumber ovum was ultrasound25min at30℃,2h for extraction process and1.5times amount of water more than material medical, and the yield of product was8.1%.3h for extraction process at30℃and10times amount of ethanol more than material medical, and the yield of product was20.3%.2、The trace elements of Fe, Ca, Mg, Zn, Se, Ge, Sr, Cu contained in lyophilized of sea cucumber ovum are good for humans’health. And the protein contents of it was61.7%.3、Comparing with blank group, the organ index in model group decreased significantly (P<0.05), however, the organ index in other groups were notably higher than model group (P<0.05).4、Comparing with blank group, the levels of estradiol and progesterone in model group decreased significantly (P<0.05), and endometrial thickness became thinner obviously (P<0.01). however, the levels of hormone and endometrial thickness in other groups were notably higher than model group (P<0.05).5、Comparing with blank group, less endometrial glands, smaller glandular, less vascular, interstitial cells were spindle-shaped, and glandular epithelium was pseudo-stratified in model group. However, more endometrial glands, larger glandular, more vascular, interstitial cells were roundness-shaped, and glandular epithelium was simple columnar in model group.6、Comparing with blank group, poorer development, fewer follicles, partially replaced by fibrous tissue, and irregular granule cells in ovarian of model group. However, well development, more follicles, occasional mature follicles, and regular granule cells in ovarian of other group.7、Comparing with blank group, the activity of SOD and GSH-Px, content of protein decreased significantly, and the content of MDA increased in ovary and uterus of model group (P<0.05). However, others were significantly improved (P<0.05). 8、 Comparing with blank group, the mortality rates and expressions of Bax and Caspase3increased significantly, and the expressions of Bcl-2decreased in ovary and uterus of model group (P<0.05). However, the other group were significantly improved (P<0.05).CONCLUSION1、Sea cucumber ovum contain both water-soluble substances and contain fat-soluble substances.2、There are lots of trace elements contained in lyophilized of sea cucumber ovum are good for humans’health, with high protein but low fat contents.3、The extracts of sea cucumber ovum can increased levels of hormone, organ index, endometrial thickness, improve ovarian morphology, protect reproductive system.4、The extracts of sea cucumber ovum can increased contents of protein, activities of SOD and GSH-Px, decreased activities of MDA, the antioxidant mechanism may be related to the inhibition of lipid peroxidation.5、The extracts of sea cucumber ovum can demote apoptosis by downregulating Bax, caspase-3expression and upregulating Bcl-2expression.
