AIM:To detect the expression of nucleobindin-2/nesfatin-1protein in gastric fundus, body, antrum and colonic mucosa of simple obese people and normal people. To analyse the distribution difference of Immune cells of nesfatin-1in different gastirc areas and between gastirc areas and colon, and the effect of NUCB2gene and NUCB2/nesfatin-1METHODS:The specimens were obtained from191obese people and normal people without chronic diseases, who were received gastro scope or entero scope examination. Divide the specimens from gastric fundus, body, antrum and colonic mucosa into groups to detect the expression of NUCB2mRNA by real-time RT-PCR. To detect the expression of NUCB2/nesfatin-1protein of normal mucosa cut off in48gasstric cancer and16colon cancer surgercal treatment by immunohistochemistry, respectively.RESULTS:The expression levels of NUCB2mRNA in gastric fundus, body and antrum of simple obese people were2.04,2.17,1.83times higher than those in corresponding tissues of normal controls (P=0.015,0.006,0.009); however, the expression level of NUCB2mRNA in colonic mucosa showed no significant difference between the two groups(P=0.069).And the expression levels of NUCB2mRNA in gastric fundus, body and antrum mucosa of normal people were2.64,2.73,1.57times higher than those in colonic mucosa(P=0.013,0.005,0.018), the expression levels of NUCB2mRNA in gastric fundus, body and antrum mucosa of simple obese people were2.76,2.79,1.74times higher than those in colonic mucosa(P=0.015,0.009,0.025); however, the expression level of NUCB2mRNA in gastric fundus and body showed no significant difference between simple obese people and normal people. The immuinohistocemical study revealed the similar distribution pattern of NUCB2/nesfatin-1in the lower1/3to2/3of the gastric mucosal glands. The expression level of NUCB2/nesfatin-1protein in gastric fundus, body and antrum tissues was significantly increased in simple obese people compared with normal people.CONCLUSION:NUCB2/nesfatin-1is widely expressed in gastrointestinal tissues of people, and the expression of NUCB2mRNA and NUCB2/nesfatin-1protein is up-regulated in simple obese people. The expression of NUCB2mRNA in gastric fundus and body is more than that in antrum.
