The active polysaccharides of authentic ingredients Poria cocos F.A.Wolf. in Yunnanhas been studied in this study. Three types of ingredients were obtained afterpolysaccharides processing by chemical and physical methods. Furthermore, chemicalcomposition, monosaccharide composition and the structure of the polysaccharides weredeeply analyzed, in which their antioxidant and anti-tumor activity were also evaluated.The results are as follows:1. Orthogonal test was used to optimize the extraction conditions of polysaccharidesfrom Poria cocos F.A.Wolf. using ultrasonic assisted hot water extraction method. Theultrasonic time, solid-liquid ratio, extraction temperature and extraction time wererespectively taken into account. The optimal extract condition of polysaccharide wasultrasonic time4h, solid-liquid ratio1:50, extraction temperature100°C and extractiontime4h.2. Alkali-soluble polysaccharides from Poria cocos F.A.Wolf. were extracted through0.5mol·L-1NaOH. Orthogonal test was used to optimize the degradation conditions usinghydrogen peroxide as degradation solvent. The optimal degradation condition was reactiontemperature90°C, reaction time4h,concentration of hydrogen10%, solid-liquid ratio1:60.3. The chemical composition and monosaccharide composition of threepolysaccharides were analyzed. Results showed that PWP were mainly composed bymannose, glucose and galactose, but PAP and PDP were both composed by glucose.Polysaccharides structure were analyzed by FT-IR, the obvious characteristic peaks ofpolysaccharide in IR spectra indicate the extraction and degradation processes are feasible.4. The antioxidant activities of PDP, including scavenging activity of DPPH radicals,hydroxyl radicals, ABTS radicals, and ferrous ions, were evaluated by established in vitrosystems. The binding curve showed the strong interaction between PDP and ct-DNA.Furthermore, DNA gel electrophoresis showed the protective effect of the PDP to DNAdamage. The antitumor activity of the polysaccharides from Poria cocos F.A.Wolf. wasstudied using human breast cancer MCF-7cell as the target, which showed that the PDPhave stronger inhibition rate on cancer cell than PWP.
