Siegesbeckiae is more commonly used herbal medicines. Three species of Siegesbeckiae (Siegesbeckiae orientalis L., Siegesbeckiae pubescens Makino, Siegesbeckiae glabrescens Makino), their aerial parts have been used as a traditional Chinese medicine. Siegesbeckiae mainly used in the treatment of rheumatism, arthrosis, detoxification, and it can also treat soreness and weakness of waist and knees, hemiplegia and other illnesses. In recent years, increasing emphasis on changes in domestic and foreign researchers studied the chemical composition of Siegesbeckiae before and after processing. This paper is to establish Siegesbeckiae and its first to ninth processed products’ HPLC characteristic peaks, as well as the study of the changes of main chemical components, Kirenol, Darutigenol and Darutoside.The main works are as follows:First, the section of literature review:The three sections are Siegesbeckiae’s chemical composition of the current research status, the study of medicines’ processing, as well as the study of the processing of Siegesbeckiae.Second, to choose Siegesbeckiae herbsIn order to establish Siegesbeckiae HPLC characteristic peaks as well as the measurement of major components,the ten regions,Fuzhou,Haozhou,Yichang,Urumqi, Beijing, Yancheng, Xi’an, Changchun,Qingdao, Changsha,we bought the Siegesbeckiae to detect, and detected stems and leaf of Siegesbeckiae.The final choice was Siegesbeckiae bought in Beijing, and we used the leaves. This is the first step of the establishment of Siegesbeckiae and its first to ninth processed products’HPLC characteristic peaks, as well as the study of the changes of main chemical components Kirenol, Darutigenol and Darutoside.Third, the establishment of Siegesbeckiae and its first to ninth processed products’ HPLC characteristic peaksWe established the HPLC characteristic peaks of Siegesbeckiae and its first to ninth processed products, and we used SunFire-C18column (4.6x150mm,5μm); the mobile phase was acetonitrile containing0.1%formic acid (A)-0.1%formic acid (B) with gradient elution; the column temperature was30℃; the flow rate was1mL/min; the detection wavelength is215nm. In this chapter the method of precision, reproducibility, sample stability were investigated, and the results were satisfactory,indicating that the precision of the method, reproducibility were well,and the samples remained stable within24hours. HPLC characteristic peaks of Siegesbeckiae are the foundation of the chemical compositions changes of Siegesbeckiae.Fourth, the measure of chemical compositions of Siegesbeckiae and its first to ninth processed productsWe take the " Chinese Pharmacopoeia"2010’s preparation of the Siegesbeckiae test solution items; the column was SunFire-C18column (4.6x150mm,5μm); the mobile phase was acetonitrile containing0.1%formic acid (A)-0.1%formic acid (B) with gradient elution; the column temperature was30℃; the flow rate was1mL/min. In this chapter the method of precision, reproducibility, sample stability,recovery were investigated, and the results were satisfactory,indicating that the precision of the method, reproducibility were well,and the samples remained stable within24hours. The result can provide a basis of changing of Siegesbeckiae’s chemical compositions for the property of this herb.In this study, we established the Siegesbeckiae and its first to ninth processed products’ HPLC characteristic peaks, and measured the chemical compositions of them. The purpose is to study the changes of chemical compositions of Siegesbeckiae before and after processing and to discuss the changes of properties and effects. Only in this way, can we use Siegesbeckiae appropriate and provide the theoretical basis and data support. It is significant for Siegesbeckiae.
