Objective:This topic is based on a TLR/NF-kappa B signaling pathways mediating inflammation to discuss the possible anti-hypertensive effect of compound tablets of Gou-teng Jiang-ya on SHR.Methods:Twenty-four SHR rats were randomly divided into three groups, each group has eight rats. There are:the model control group(SHR), compound tablets of Gou-teng Jiang-ya treating group(SHR-G) and valsartan treating tablets(SHR-D). And another eight WKY rats are used as blank control group(WKY). All rats were administered daily gavage for6weeks and gavage volume is lOmL/kg, SHR’s and WKY’s rats are administered directly with distilled water; SHR-G group:Rats are administered with compound tablets of Gou-teng Jiang-ya diluted with distilled water to a concentration of43.2mg/mL;SHR-D group:Rats are administered with valsartan diluted with distilled water to a concentration of0.72mg/ml. Blood pressure is measured by Caudal artery manometry before gavage, the first7day, the14th, the first21, the first28, the first35and the first42after gavage.The mean value of each group’s blood pressure at each time point is counted up to explore the effect of compound tablets of Gou-teng Jiang-ya on the blood pressure of SHR’s rats.Plasma and thoracic aorta specimens were collected after6weeks’ gavage to detect the concentration of AngⅡ, MCP-1, Ang(1-7) in the blood of rats by ELISA and to detect the protein level of TLR4and NF-κBp65in thoracic aorta using immunohistochemical method.So that,we can further explore the effect of compound tablets of Gou-teng Jiang-ya on serum inflammatory factors AngII,MCP-1,Ang(1-7) and on the protein level of TLR4and NF-KBp65which belong to TLR/NF-κB signaling pathway, aiming to know whether compound tablets of Gou-teng Jiang-ya effects blood pressure of SHR’s rats through affecting nflammatory reaction mediated by TLR/NF-κB signaling pathway.Results:(1) Blood pressure of SHR’s rats is significantly higher than that of WKY’rats; The blood pressure of SHR-D and SHR-G significantly decreased after treatment of valsartan tablets(positive control) and compound tablets of Gou-teng Jiang-ya,and maintain close to that of WKY’s rats after5or6week’s treatment.(2) The plasma concentrations of AngⅡ and MCP-1of SHR rats are significantly higher than WHY rats; The concentrations of AngⅡ and MCP-1in SHR-D and SHR-G significantly decreased after treatment of valsartan tablets and compound tablets of Gou-teng Jiang-ya,however the concentration Ang(1-7) is just the opposite: The plasma concentrations of Ang(1-7) of SHR rats are significantly lower than WHY rats; The concentrations of Ang(1-7) in SHR-D and SHR-G significantly increase after treatment of valsartan tablets and compound tablets of Gou-teng Jiang-ya(3) The protein level of TLR4and NF-KBp65in thoracic aorta of SHR rats are significantly higher than WHY rats. The protein level of TLR4and NF-κBp65in thoracic aorta of SHR-D and SHR-G significantly reduced after treatment of valsartan tablets and compound tablets of Gou-teng Jiang-ya.Conclusion:(1): Compound tablets of Gou-teng Jiang-ya can significantly reduce SHR rats’blood pressure.(2): Compound tablets of Gou-teng Jiang-ya can reduce SHR rats’ concentrations of proinflammatory cytokines Ang II and MCP-1, and increase that of anti-inflammatory cytokines Ang (1-7).(3): Compound tablets of Gou-teng Jiang-ya can inhibit the expression of pro-inflammatory cytokines TLR4and NF-κB p65inSHR rats’thoracic aorta.In summary the following conclusions can be drawn:Compound tablets of Gou-teng Jiang-ya may inhibit the expression of pro-inflammatory cytokines and increase that of anti-inflammatory cytokines through TLR/NF-κB signaling to achieve the anti-hypertensive function.
