GBPs (Glycan-binding proteins) which could recognize and specifically bind to intracorporal carbohydrate, play an important role in various fundamental biological processes, including cell recognition, signal transmission, cell growth, differentiation, apoptosis and migration, et al. It has been confirmed that GBSs are closely related to development of several types of cancer.Current studies show that, the process of HB (Hepatitis B), HC (Hepatic Cirrhosis) and HCC (Hepatocellular carcinoma), are accompanied by alteration of GBPs in the serum and tumor tissues of patients.Whether there are a similar changes in the saliva is still not clear.In our study, carbohydrate microarray was used for investigation of GBPs in the saliva from HB,HC and HCCpatients. We select the carbohydrate probes which could recognise and specifically binding to the common and special GBPs in the saliva from HB, HC and HCCpatients,provide experimental basis for early diagnosis of liver cancer, and study the relevance between investigation of GBPs in the saliva and the process of HB, HC and HCC. The experiment contains two parts:(1) We usedcarbohydrate microarrays (contains41carbohydrate probes) to probe the alteration of GBPs in the saliva from HB,HC and HCCpatients. As a result, in the saliva of HBpatients,11carbohydrate robes (e. g. Man, Gal and NeuAc) showed increasingly signals, while8robes (e. g.Man-3Glycan,6’-SL and iso-A-Pentasaccharide) showed decreased signals; in the saliva of HCpatients,8robes (e. g. Man,Gal and NeuAc) showed increasingly signals, while10robes (e. g.3-SNL,NA2F and LNDF I) showed decreased signals; in the saliva of HCCpatients,13robes (e. g. Gal, GalNAc and Hyaluronic acid) showed increasingly signals, while7robes (e. g.Xyl, Man-3Glycan and NGA2Glycan) showed decreased signals.The result showed that,6robes (e. g. NA2F, LNDF I and Lewis-Y tetrasaccharide) could recognize the GBPs which differentially expressed in the whole process of HB,HC and HCC;8robes (e. g. Man, Man-9Glycan and N-NeuAc) could recognize the GBPs which had characteristic expression in HCC;9robes (e. g.6a-Mannobiose,3’-Sialyl-Lewis-a tetrasaccharide and NA2F) showed increasingly signalsfrom HB to HC,while6robes (e. g. 3a-Fucosyl-N-acetylglucosamine,xylose and Lactose F) showed increasingly signalsfrom HC toHB.(2)Based on the result of carbohydrate microarray,we designed a saliva microarraytovalidate the specific GBPs of HB,HC and HCC, it showed same result as carbohydrate microarray.
