The theory of "The Lung Being Relative to the Large intestine" originated from "Huangdi Neijing" reflects the holistic view of Chinese Medicine on organs. As a specific form of the Lung Being Relative to the Large intestine Theory, it is being improved and tested in the development and clinical application of traditional Chinese Medicine.Chronic obstructive pulmonary disease (COPD) is a sort of lung disease which is characterized by on fully airflow limited. COPD mainly results form a long term infection and inhaling much more harmful substances in daily life, as well as the decrease of resistance capacity for disease and Physical conditions. COPD is a serious disease which causes the forth mortality worldwide. Our research is based on the theory of "the Lung Being Relative to the Large intestine" originated from traditional Chinese Medicine, and by establishing the COPD rats model, using XuanBaiChengQiTang and its separated parts to investigate the mechanism of relaxing large intestine to treat COPD from the angle of impacting the variety of neuropeptide. The thesis is totally divided into three parts:Part I SummaryThis section describes the research progress of neuropeptide SP and VIP and the status of traditional Chinese medicine on treating chronic obstructive pulmonary disease (COPD). Modern studies have found that neuropeptide SP and VIP play important roles in the respiratory diseases, not only in the regulation of respiratory function but also in the respiratory system inflammation of neurogenic. Traditional Chinese medicine has earned much more progress by "overviewing the whole body" and using multi-ways and multi-angle treatment to treat COPD.Part II Theory investigationWe attempt to investigate the therapy of relaxing the large intestine on treating COPD by combining modern study on the theory of "the Lung Being Relative to the Large intestine" with that noted in "Shang Han Lun" formulas in and further understand the thought of Zhang Zhongjing about "the Lung Being Relative to the Large intestine."Besides, we meat to inherit and carry the thinking of XuanBaiChengQiTang which could perform the effectiveness of treating lung disease by treating intestine noted in "Shang Han Lun" forward. All the investigation would help to demonstrate the application and initially discover the mechanism of using XuanBaiChengQiTang which had the effectiveness of vent heat from organs recorded in Shang Han Lun for treating COPD.Part Ⅱ Trial processingObjective: To explore the principle of "relaxing the large intestine" on treating COPD, we established model COPD of rat and used XuanBaiChengQiTang and its separated parts as the administration drugs. The subjects were divided into five groups:blank group, model group, intestine treating group, lung treating group, combination treating of intestine and lung group. By detecting the performance of behavior of rats and the pathology changes on lung, we meat to study the treatment of XuanBaiChengQiTang and its separated parts. Immunohistochemistry method and Enzyme-linked immunosorbent assay method were taken to test the changes of amount on neuropeptide SP and VIP in order to study the adjustment effect of XuanBaiChengQiTang as well as its separated parts on COPD rat model. Besides, we expected to find the relationship between drugs treatment and the variety of neuropeptide SP and VIP. Our study was mainly designed to recover the efficacy mechanism of treating COPD via "treating form intestine" method on the angle of impacting neuropeptide changes.MethodThe model of COPD rats were established by being injected lipopolysaccharide meanwhile given cigarettes fume atmosphere. After that, all the subjects were divided into five groups including blank group, model group, intestine treating group, lung treating group, combination treating of intestine and lung group. XuanBaiChengQiTang and its separated parts were chosen as the administration drugs and the blank group was administration with pure water as well as model group. The dosage of drugs and pure water was equivalent and the time zone was7days. We recorded the performance of every rats from every group in order to inspect the effects of our drugs. HE staining was used to find the morphological changes of the lung of rats by observing under optical microscope. Immunohistochemistry method was taken to find the status of amount and dispersion of neuropeptide SP and VIP. Enzyme-linked immunosorbent assay (ELIS A) was used to measure SP and VIP levels in lung homogenates.ResultBehavior performance:Compared to blank group, other groups show the changes from dysphoria to inactive. As the processing of our trial, the fur of these animals grew withered, yellow, dull or even fall off. Furthermore, shortness of breath, wheezing, abdominal breathing became heavier and the red status on their nose was more serious with more secretions, as well as sneezing lung.Conditions of drink increasing, activity decreasing, crouching and dry stool were showed in the process. Weight loss in the model rats was more serious compared with blank group.Pathological observation:The results of observation under optical microscope showed that blank group had no abnormal conditions about lung; compared with the blank group, bronchial epithelial necrosis and cilia loss were found in the model groups. There had shedding of epithelial and other substances within the lumen, and a small amount of inflammatory cells infiltration was showed on the wall of vessel. Alveolar structural disorder, partly fused into alveolar bulla as well as severe alveolar vascular congestion. Compared with the model group:there was a release in the lung tissue of intestine group. Compared with lung treatment Group:combination treating of intestine and lung group showed better recovery on lung.Immunohistochemistry:Neuropeptide SP was generated in lung bronchial epithelial tissue and pulmonary interstitial tissue. And model group showed much more expression than blank group. Compared with the model group, the intestine treatment group and the combination treatment of lung and intestine group showed decrease in the expression of SP. Neuropeptide VIP expression on the bronchial epithelial tissue in blank group rats grew stronger as contrary to significantly decreasing in the model group. Neuropeptide VIP was generated more in every administration groups.Enzyme-linked immunosorbent assay showed that compared with control group, SP was increased in lung tissue in model group while VIP were decreased significantly (P<0.01). Compared with model group, SP was lower in lung group, intestine group and lung-intestine group. VIP was increased significantly (P<0.01or P<0.05). Compared with lung group, SP was reduced significantly in lung-intestine group (P<0.05).Conclusion:In summary, both XuanBaiChengQiTang and its separated parts could improve the effectiveness of treating lung pathological changes of the COPD rat models by the therapy of relaxing the large intestine or relaxing the large intestine. Besides, they can adjust the contents of Neuropeptide SP and VIP in the lung tissue of COPD model rat.The expression of SP and VIP were regulated by the therapy of relaxing the large intestine or relaxing the large intestine on the base of treating lung disease. This may be one of the mechanisms of why treating from intestine is effect on COPD.
