Background:Cardiovascular disease and cerebrovascular disease is a common disease hazard to human health. According to Health Ministry statistics, the amount of people who afflicted with cardiovascular disease grows700million a year. more than150million existing patients, It gives a heavy burden to families and society. In recent years, the occurrence of cardiovascular events showed a young trend. Atherosclerosis is an important factor in cardiovascular disease. Clinical research and basic research by scholars, and from the whole level to organ level, from the molecular level to the cellular level, Revealed the fact that atherosclerosis is an important role in cardiovascular and cerebrovascular diseases. Today, atherosclerosis is an inflammatory disease has reached a consensus. Moreover, From this perspective to treat atherosclerosis can make atherosclerotic plaque stability increased. Research the factors which enables atherosclerotic plaque stability increases, it is important for the prevention of acute onset and always without aura of cardiovascular and cerebrovascular events. With its unique treatment system, Traditional Chinese Medicine Played a pivotal role in the reversal of atherosclerosis and in the vulnerable plaque regression.Objective:Through animal experiments to investigate the impact of Chinese medicinal with actions of regulating blood lipid and invigorating pulse-beat on atherosclerotic rabbit model lipid levels, and the effect on AS plaque stabilizing, As well as including its impact on inflammatory factors. Dynamic observation the efficacy and mechanism of action of Chinese medicinal with actions of regulating blood lipid and invigorating pulse-beat. Provide experimental and theoretical basis for the prevention and treatment of early atherosclerosis.Methods:The rabbit model of atherosclerosis was prepared by the high fat diet and balloon injury method, they were randomly divided into five groups, normal group, model group, sham-operation group,atorvastatin group (Referred to as western medicine group,1.1mg/kg/d). regulating blood lipid and invigorating pulse-beat group (Referred to as Chinese medicine group,7.3g/kg/d) Treatment groups were treated by gavage. The6weeks,12weeks after treatment were select as the observation points. at two points perform ultrasonic testing. Blood samples are drawn from auricular veins to determine total cholesteroI(TC), triglyceride(TG), low-density lipoprotcin(LDL-C).After6weeks、12weeks kill the animals and removed the material. Hematoxylin-eosinstaining dyeing. Pathological arterial tissue was observed by electron microscopy. The serum C-reactive protein (CRP), transforming growth factor-β (TGF-β), interleukin6,10,17(IL-6, IL-10,IL-17) levels were detected by enzyme-linked immunosorbent assay (ELISA). mRNA expression of inflammatory cytokines detected by RT-PCR. Evaluate the impact of Chinese medicinal with actions of regulating blood lipid and invigorating pulse-beat on atherosclerosis in rabbit model Comprehensively.Results:(1):Atorvastatin group and Regulating blood lipid and invigorating pulse-beat group could significantly reduce atherosclerotic rabbit model of serum cholesterol, low-density lipoprotein(P<0.05).(2):Pathological arterial tissue:After6weeks and12weeks time points, Model group and sham-operation group can see the plaque formation. Compared with the model group, atorvastatin group and Regulating blood lipid and invigorating pulse-beat group can significantly reduce arterial intimal injury, under the microscope, can only see small amount of lipid calm.(3):The results of serum ELISA:The serum CRP, IL-6, IL-17of model rabbits on different time points were significantly higher (P<0.01), and the serum TGF-β, IL-10were decreased significantly than those of the sham-operation group (P<0.01). The serum IL-6,CRP,IL-17levels of regulating blood lipid and invigorating pulse-beat group decreased significantly (P<0.01), the serum TGF-β, IL-10levels were increased significantly than those of the model group (P <0.01), and the differences between regulating blood lipid and invigorating pulse-beat group and the atorvastatin group were not significant.(4):The results of RT-PCR:Compared with sham-operation group, the gene expression of CRP、IL-6、IL-17in model groups were significantly higher, there is a significant di fference (P<0.05). Compared with model group, The gene expression of CRP、IL-6、IL-17in regulating blood lipid and invigorating pulse-beat group and atorvastatin group were decreased significantly (P<0.05). Compared with the sham-operation group, model group arterial tissue TGF-β、 IL-10mRNA levels were decreased, the difference was significant (P<0.05). Compared with model group, atorvastatin group and regulating blood lipid and invigorating pulse-beat group arterial tissue TGF-β, IL-10mRNA levels were significantly increased (P<0.05).Conclusion:1:Chinese medicinal with actions of regulating blood lipid and invigorating pulse-beat can regulate fat metabolism, It also has a certain role in lowering Lipid in rabbits model with atherosclerosis.2:Chinese medicinal with actions of regulating blood lipid and invigorating pulse-beat has anti-atherosclerotic effect, It also has a significant effect on the stability of atherosclerotic plaques. Enables atherosclerotic plaque rupture tends to decrease. Reduce acute cardiovascular and cerebrovascular events caused by plaque instability risk.3:Chinese medicinal with actions of regulating blood lipid and invigorating pulse-beat can affect the expression of lipid-related inflammatory mediators, reduce inflammation, inhibit arterial intima. Inhibit the expression of inflammatory factors(CRP、IL-6、IL-17) within the plaque, Play a important role in anti-atherosclerotic.
