Objective:This study intended to rat focal cerebral ischemia model (MCAO) as the observation object, observing the curative heart meridian, the pericardium meridian of neurologic deficits in rats, cerebral infarction volume, microvascular density and serum of rats and the effect of TGF-beta expression in the brain tissue. To explore the curative heart meridian, the pericardium meridian intervention effect of cerebral ischemia injury and nerve protection, the role of angiogenesis, as the heart meridian, the pericardium meridian treating cerebrovascular disease and from heart to treat brain research provides the experimental basis.Methods:Adopt the method of quadratic random60male SD rats were divided into:normal group, model group, control group, the heart sutra, the pericardium6groups by the group, the lung group, each group of10, only to tie or bundled+acupuncture treatment respectively. In the building after2h,6h,24h,48h,72h lines of nerve function defect grade and cupping treatment was carried out on the acupuncture point group. Rats drawn by TTC staining method to detect after cerebral infarction volume; Under a microscope measuring brain microvascular density; Double resistant sandwich enzyme-linked immunosorbent (ELISA) method to detect the serum content of TGF-beta in; Immunohistochemical staining(SABC)method to detect infarct area level of expression of TGF-beta in the brain tissue.Result:1. The normal group, control group without neural function defect scale; Building after each time point, model group, the heart meridian, the pericardium meridian group and neural function defect scale, lung meridian group was obviously higher than that of normal group and control group, the difference is statistically significant (p<0.01),2h each building module when the score is no statistical difference (p>0.05); Group compared with model group, heart sutra in48h,72h neural function defect scale is reduced, the difference was statistically significant (p<0.01,p<0.05), pericardial by groups within24h,48h,72h neural function defect scale is reduced, the difference was statistically significant(p<0.05,p<0.05,p<0.01), lung meridian at various time points score had no statistical significance; Heart sutra, the pericardium group, the lung group each time point score differences had no statistical significance (p>0.05); Compared with2h, the heart sutra group at24h,48h,72h score difference was statistically significant (p<0.05), the pericardium meridian group in72when the difference was statistically significant(p<0.05), model group, the lung group at various time points score difference had no statistical significance.2. Each rat whole brain volume was no difference (p>0.05); Compared with normal group and control group, model group, the heart meridian, the pericardium meridian group, pulmonary infarction, infarct volume rate of acupuncture point group was obviously increased (p<0.01); Compared with model group, the heart meridian, the pericardium meridian group and pulmonary infarction, infarct volume rate of acupuncture point group was obviously decreased (p<0.01); Compared with lung meridian group, heart meridian, the pericardium meridian group infarction and cerebral infarction volume rate has dropped, but had no obvious statistical difference (p>0.05)3. Model group, the heart meridian, the pericardium meridian group, lung meridian group of cerebral infarction area in the microvascular density were higher than in normal group, control group, the difference was statistically significant (p<0.01); Heart meridian, the pericardium meridian group, cerebral infarction area in the microvascular density of lung meridian group was obviously higher than that of model group, the difference was statistically significant (p<0.01); Heart meridian, the pericardium meridian group cerebral infarction area in the microvascular density higher than that of lung meridian heart meridian group, the difference was statistically significant (p<0.01); Heart meridian, the pericardium meridian group cerebral infarction area in the microvascular density has no obvious difference (p>0.05)4. The normal group, control group of serum concentration of TGF-beta differences had no statistical significance (p>0.05); Model group, the heart meridian, the pericardium meridian group TGF-beta concentration in serum, lung meridian group were higher than in normal group, control group, the difference is statistically significant (p<0.01); Heart meridian, the pericardium meridian group, lung meridian group TGF-beta in serum concentrations higher than the model group, significant differences (p<0.01); Heart meridian, the pericardium meridian group TGF-beta concentration in serum is higher than the lung meridian group, the difference is statistically significant (p<0.01); Pericardium heart sutra group by group has no statistically significant difference (p>0.05)5. The normal group, control group of TGF-beta expression differences in brain tissue had no statistical significance(p>0.05); And normal group, control group than in brain tissue of lung meridian group TGF-beta expression increased (p<0.05), model group, the heart meridian, the pericardium meridian group brain TGF-beta expression significantly increased(p<0.01); Than with the model group, the pericardium meridian group brain TGF-beta expression increased obviously (p<0.01); Than to the lung meridian group, brain tissue TGF-beta expression increases heart meridian group (p<0.05), the pericardium meridian group brain TGF-beta high expression(p<0.01); Heart meridian and the pericardium meridian group, cerebral infarction area TGF-beta expression increased (p<0.01)Conclusion:1. Cupping pericardium meridian group have role to improve the neural function defect scale.2. Cupping heart meridian, the pericardium meridian, lung meridian can reduce cerebellar infarction volume.3. Cupping heart meridian, the pericardium meridian can increase the expression of TGF-beta in serum and brain tissue, and effective treatment for cerebral ischemia injury.4. Cupping heart meridian, the pericardium meridian can increase MCAO rats microvascular density of infarction area.
