| Purpose：By copying subcutaneous injection of nitroglycerin migraine ratsmodels, observed the effect of acupuncture on migraine rats behavior change andthe changes of plasma SP and CGRP level, discusses acupuncture mechanism ofprevention and treatment of migraine.Materials and methods:1.50healthy male SD rats, weight is about250+50g, by liaoning university of traditional Chinese medicine biotechnologylab. Adaptive feed a week after the experiment. Rats can be divided into5groupsaccording to the completely random method (blank control group, model controlgroup, preventive acupuncture group, acupuncture group, acupuncture prevention+treatment group),10in each group. Blank control group: normal breeding,7days without any processing; Model control group: normal7days,8days ofsubcutaneous injection of nitroglycerin building; Acupuncture prevention groups:acupuncture treatment for7days,20-30min/d, day8subcutaneous injection ofnitroglycerin building; Acupuncture treatment group: normal7days,8days ofsubcutaneous injection of nitroglycerin to build mold, after the success of thebuilding, acupuncture20-30min; Acupuncture prevention+therapy groups:acupuncture treatment for7days,20-30min/d, day8subcutaneous injection ofnitroglycerin to build mold, after the success of the building, acupuncture20-30min.2. The subcutaneous injection of nitroglycerin (10mg/kg) preparation ofmigraine model rat.3. The method of acupuncture: find out: best, gb34(bilateral), lr3(bilateral),close (bilateral), acupuncture20min.(1) the will, the center of the parietalbone,2mm forward or backward inclined.,(2) gb34: knee posterolateral fibularentered below cavity, stab6mm.(3). Lr3: hind leg foot back cavity betweenfirst and second metatarsal, stab to1mm. Forelimb:(4), the lateral, is apartfrom the wrist ulnar radial between about3mm. stab2mm..4. Observe behavioural changes of rats after building, using the following three as observation indexes:(1) in the rat ear red duration: observe and record ratsears red appear and disappear time points and calculate its duration;(2) inthe rat forelimb scratching his head frequently behavior duration: frequentobservation and record rat forelimb scratching his head scratching his head morethan5times in a row (as standard) behavior in time and disappear (continuousscratching his head number less than5times) point in time, and calculate itsduration;(3) minutes time statistics rats scratching his head: in30minutesfor a period of time, a timebox records since the building after31minutes to180minutes of the total of scratching his head.5. Using enzyme-linked immunosorbent (ELISA) method to determine the serum ofrats in the change of SP and CGRP content.Results:1. The model control group and about4min after subcutaneous injection ofnitroglycerin or ear red, forelimb scratching his head frequently, symptoms,the symptoms ranging from170-180minutes. Groups compared with the period withthe blank control group shown in each time period have frequent forelimbsscratching his head number increase symptoms (P<0.05) indicating buildingsuccess.2. Acupuncture effect on behavioral changes of rats: ear red, forelimb frequentmonths duration for statistics can be found that acupuncture prevention group,the acupuncture treatment group, acupuncture prevention+treatment group caneffectively reduce the frequent ear red, forelimb scratching his head symptomduration, compared with model control group was statistically significant (P<0.05); In30min for a period of time, period statistics31-180min in frequentissue number found in rats, acupuncture prevention group, the acupuncturetreatment group, acupuncture prevention+treatment group at each time all caneffectively reduce the number of scratching his head, compared with same periodmodel control group P<0.05, acupuncture prevent+treatment group andacupuncture group, acupuncture treatment group to compare curative effect is better (P<0.05).3. Acupuncture serum of rats in SP, CGRP content change: the influence ofacupuncture prevention group compared with model control group: SP, CGRP contentdecreased, prove that acupuncture has prevention effect of migraine rats (P<0.05); Acupuncture treatment group compared with model control group: SP, CGRPcontent decreased, prove that acupuncture has a therapeutic effect on migrainerats (P<0.05); Acupuncture prevent+treatment group and acupuncture group,acupuncture treatment group comparison: cut down the content of SP, CGRP andprove that acupuncture+the curative effect of treatment group was better thantreatment and simple preventive alone (P<0.05).Conclusion:1With reference to Cristina method, successfully cloned type nitroglycerinmigraine rats model.2. Acupuncture can effectively improve the migraine rats behavior change, withacupuncture prevention+treatment group curative effect significantly.Type3. Nitroglycerin migraine rats model, SP, CGRP content in serum of migrainepathogenesis and SP mediated hypersensitivity and CGRP blood vessels caused byexcessive expansion and neurogenic inflammation.4. Acupuncture can effectively reduce the migraine rats in serum, SP, CGRPcontent in to acupuncture prevention+treatment group effect obviously.5. Acupuncture can through reduce the migraine rats SP, CGRP content in serum,and alleviate cerebral over-expansion and hypersensitivity, adjust the cerebralblood flow, reduce the neurogenic inflammation, and to have prevention andtherapeutic effect on migraine. |
