PART ONE THE DESIGN OF WEAK ALKALINECANCER-INHIBITING LIQUIDObjective: Cancer patients often suffer from cancer relapse andmetastasis after treatment despite of the recent treatment advancement. Thus,discovering a novel reasonable therapy for cancer is a matter of greaturgency. Here, we designed a Weak alkaline cancer-inhibiting liquid(WACIL).Methods: Making WACIL in different mass ratio of K andNa(mk/mNa)with NaHCO3, KCl and ddH2O and the total volume is500mL, then pH values was determined.Results:When mk/mNais6(4g NaHCO3and24g KCl), the pHvalue of the500mL WACIL is near to7.45.Conclusion: WACIL is made of4g NaHCO3and24g KCl(mk/mNa=6) with the total volume of500mL by adding some ddH2O,then filtered after fully dissolved. PART TWO IN-VITRO EXPERIMENTS OF WACILREGULATING CANCER CELLS’GROWTH,METASTASIS AND REGRESSIONObjective: Observe wether WACIL could regulate the growth,metastasis and regression of cancer cells, and explore the probableregulating machanism.Methods: In-vitro experiments were used to observe the impact ofWACIL to the growth, proliferation, cell cycle, apoptosis, invasion andmetastasis of the three cultured cell lines: cervical aqueous cell carcinomaSiHa cells, non-small cell lung carcinoma H1299cells and human breastcarcinoma MDA-MB-231cells.Results:WACIL significantly repressed the growth and proliferationof the three cell lines: G2/M phase cells were increased indicating cell cyclewas arrested in G2/M phase. Cells became round, accompanying withnucleuses concentrated and casease-3activity enhanced as well asmitochondrial membrane potential decreased suggesting cell apoptosis thenumber of living cells were reduced and membranes were ruptured, theregression phenomenon was observed and cancer cell migration wassignificantly inhibited (P<0.001). The mRNA expression of SCCA1,cyclinB1, MMP-2and MMP-9were down-regulated while Casease-3mRNA up-regulated; and the protein expression of SCCA1, cyclinB1, MMP-2were down-regulated, while casease-3protein up-regulated.Conclusion: WACIL could regulate cancer cell’s growth, metastasisand regression. WACIL could repress the growth and metastasis of cancercells. More importantly, this study systematically analyzed the interactivemechanism between WACIL and cancer cell itself along with its pHmicro-environment, which uncover the importance of changing the tumormicro-environment. PART THREE IN-VIVO EXPERIMENTS OF WACIL’sINFLUENCE ON THE GROWTH AND METASTASIS OFTRANSPLANTATION TUMORObjective: To study the influence of WACIL on the growth andmetastasis of cancer cells and explore the potential treatment significance.Methods: Established the nape subcutaneous transplantation model ofMDA-MB-231cell in nude mice to investigate the effect of WACIL on thegrowth and metastasis of subcutaneous tumor.Results:In-vivo studies suggested that tumor growth rate wassignificantly slower in the WACIL-treated group than that in the controlgroup; lung tissue of the WACIL-treated group didn’t show cancermetastasis by HE staining, other than the control group.Additionally, WACIL could degrade tumor angiogenesis.Conclusion: WACIL could control the growth of cancer cells andinhibit metastasis, showing potential preventing and treating significanceto tumor.
