Purpose:To observe the Qijishenkang Granule for IgA Nephropathy (IgAN, IgANephropathy) in the rat kidney urine red blood cell count,24hours urine proteinquantitatively and kidney tissue IL-4mRNA, IL-13mRNA expression and theinfluence of circulating immune complexes (CIC) from serum concentrations, andexplore the mechanism of Qijishenkang Granule in the treatment of IgAnephropathy.Material and method:Methods58SD rats were divided randomly into blankgroup, model group, Qijishenkang Granule low dose group,middle dose group andhigh dose group, telmisartan group. The rats were given oral bovine serum albuminacidified water and tail vein injection of lipopolysaccharide and subcutaneousinjection of carbon tetrachloride, composite process of castor oil preparationof experimental IgA nephropathy rat model. In the eighth week, the normal group,model group rats were given equal volume of distilled water, the rest of therats treated with corresponding drugs orally,1times a day, for5weeks, atthe twelfth weekend, Fasting, but the water,24h urine samples were collectedin metabolic cages, detection of urinary red blood cell count,24hour urinaryprotein quantitative, Abdominal aorta taking blood, detection of serumcreatinine, urea nitrogen, by using enzyme-linked immunosorbent (ELISA) todetect the contents of serum circulating immune complex. In kidney tissue byreal-time fluorescent quantitative PCR method to detect IL-4mrna and IL-13mrna expression.Results:1.Qijishenkang Granule in each dose group compared with the model group, urinered blood cell count decrease,24hours urinary protein quantitative decreasedsignificantly (P<0.01), the treatment groups showed no significant differenceto each other(P>0.05). 2.Compared with the model group: Qijishenkang Granule middle dose group and highdose group the expression of IL-4and IL-13mRNA had expression decreasedsignificantly (P<0.01), Qijishenkang Granule in each dosage group circulatingimmune complexes from serum concentrations were decreased significant withdifferences (low and high does group,P<0.01,middle dose group, P <0.01).Withtelmisartan group comparison: Qijishenkang Granule middle dose group and highdose group the expression of IL-4and IL-13mRNA had significantdifference(P<0.01), the dose of Chinese medicine serum circulating immunecomplexes concentrations had no significant difference(P>0.05).No significantdifference in each dose group of Chinese medicine serum concentration ofcirculating immune complexes (P>0.05).Conclusion:1. Qijishenkang Granule can reduce the IgA nephropathy rat urine red blood cellcount, reduce the24h urine protein quantitative. Every agent was stilbenethistle renal granular IgA nephropathy has certain therapeutic effect onexperimental rats.2. Qijishenkang Granule have adjustment of IgA nephropathy rats kidney IL4,IL-13mrna expression and the role of serum concentration of serum circulatingimmune complexes.
