Florfenicol is a third generation product of chloramphenicol.It is a kind of new broad-spectrum antibiotics for animal. Because of its broad antibacterial spectrum, good absorption, wide distribution, safe and efficient characteristics, especially not induced aplastic anemia occurred,florfenicol has been widely used in the treatment of bacterial diseases in aquaculture, livestock, poultry breeding.It has a good effect in the treatment of porcine contagious pleuropneumonia,porcine pasteurellosisn, Streptococcus, Haemophilus parasuis disease, asthma and other respiratory diseases of poultry and pigs. Scutellaria baicalensis was one of the representatives of the Chinese herbal medicine and it was used to treat many disease, baicalin as the main component. As the main drug prescriptions in Scutellaria baicalensis Georgi plays an important role in the treatment of animal diseases, especially in Shuanghuanglian oral liquid and florfenicol powder are often used in combination to control the animal respiratory tract infection, received good results.However, Scutellaria and florfenicol combined use in animal,the pharmacokinetic changes or not, has not yet been studied. The influence of baicalin on florfenicol dynamic in the chicken was studied in this paper. The results would provide a useful reference for the rational use of traditional Chinese and Western medicine, scientific compatibility.In this experiment,30Daheng broilers were randomly divided into A, B groups (n=15). A group:single dose oral gavage of florfenicol; group B:florfenicol and baicalin solution of single dose at the same time gavage oral administration. Florfenicol was administered at a dose of30mg/kg.b.w, baicalin dosage is10mg/kg.b.w. After administration,0.5ml blood of chickens in A, B groups were collected from wing vein blood at0.083,0.167,0.333,0.5,0.75,1,2,4,6,8,10,12,24h,and plasma was extracted.High performance liquid chromatography method was used for the determination of blood drug concentration.High performance liquid chromatography method was used for the determination of blood drug concentration.Standard curve regression equation is:y=21627x+5218, the correlation coefficient R2=0.9958, florfenicol at the concentration of0.1~20μg/mL, linear range and good relationship between peak area. According to the signal-to-noise ratio of S/N=3computing the minimum detection limit is0.03μg/mL, S/N=10quantitative calculation of limit is0.1μg/mL.The pharmacokinetic parameters were analized by DAS2.0pharmacokinetic analysis programming and data was analized by SPSS (17). The following were the main pharmacokinetic parameters:Cmax were(4.2433±0.206mg/L) and (5.2845±0.180mg/L), Tmax were1.933±0.258h hour and0.917±0.122h hour, AUC (0-∞) value were27.519±1.346mg/L*h and19.93±0.812mg/L*h, MRT (0-∞) were6.727±0.103hours and3.512±0.012hours, t1/2z were3.262±0.161hours and2.453±0.041hours, CLz/F were1.09±0.156L/h/kg and1.505±0.127L/h/kg, Vz/F were5.132±0.465L/kg and5.329±0.386L/kg. And florfenicol pharmacokinetic parameters were compared, the combination group of chicken Cmax (P<0.05) increased significantly, Tmax advanced significantly (P<0.01), but the AUC value (P<0.01), MRT (P <0.01) and t1/2z (P<0.05) were significantly smaller, CLz/F (P<0.05) significantly faster, Vz/F had no significant change (P>0.05).The results show that, under the same route of administration of baicalin has a greater influence on the drug in vivo pharmacokinetics of florfenicol in chicken. It is concluded that baicalin can speed the absorption of florfenicol and accelerate its elimination in chickens.
