Based on the traditional Chinese medicine theory, Huanglian Zhusan Keli(HLZK) is a advanced decoction pieces which keeps medicine property just with physical changes, in order to save decoction time and resource of Coptidis Rhizoma. According to it’s own characteristic, had done a series study on preparation, application of decoction processing, building quality standards, contrastive study with traditional ones in chemical aspect and fingerprint study, to complete pharmacy researches and build foundation for further study and factorial production.Taking appearance of zhusan keli and difficult or not of shaping as the evaluation index, the preparation research was studied with method of mono-factorial test. The final process is that the Coptidis Rhizoma powder which has passed NO.7drug sieve was mixed water by9～11ml/10g, shaping, and drying it under80℃for3hours.The study of decoction process of zhusan keli, decoction efficiency curve was described to estimate how decoction time affect it with mono-factorial test. Yield of dry extract and the content of berberine hydrochloride were set as dependent variables, optimizing the process by central composite design-response surface methodology (CCD-RSM). The better methods of decoction process as follow:adding water15times as zhusan keli, no immersing process, decoction2times, and every time of15minutes.Setting yield of dry extract, the content of extracts and berberine hydrochloride as dependent variables, from the chemical level, used multi-target grading method to study the relationship between zhusan keli and the traditional one. The final result was that1gram of zhusan keli was equal to1.780gram traditional one just in chemical dosage.According to the guidance of the quality standards of Coptidis Rhizoma in Chinese Pharmacopoeia2010edition1st part, this part of paper had finished a whole quality standards of Huanglian zhusan keli. HPLC method had been adopted to control the content of alkaloids. The determination of berberine hydrochloride was required to be not less than5.0%, the sum of the epiberberine, coptisine and palmatine were required to be not less than3.3%.In the fingerprint study, HPLC method was used to establish common patterns from respective of zhusan keli and traditional slice one. The similatity was greater than0.995with9common peak in both of them.The research has showed that preparation is reasonable and practicable, application condition better than slice, the quality standard is completed, and improved the advantage of zhusan keli on efficiency especially in saving processing time and resource of Coptidis Rhizoma by multi-target analysis of chemical content.
