Hemoglobin is an important raw materials for the preparation of HBOCS, and its main function is carrying and releasing oxygen. But studies have shown that the hemoglobin in the blood is a potential toxic molecule and may induce the oxidative stress when it release from red blood cell sin the situation of tissue in juryor inflammation. Studies have shown that hemoglobin has the similar function with peroxidase in consuming H2O2. But at the same time hemoglobin could generate a large number of reactive oxygen species (ROS) and activity of nitrogen (RNS). ROS/RNS damage to the body by itself. And are more likely to be ROS/RNS oxidation Hb reaction, formed to the Fe3+Hb (without the function of carry oxygen) and at the same time induce to the generation of reactive oxygen free radical. Hb constantly be oxidated to Fe4+Hb, accumulate a mass of oxygen free radicals at same time, Fe4+Hb is a strong oxidizer, which cause lipid peroxidation, DNA fracture and so on a series of cascade.Hemoglobin oxidative stress mainly focuses on adding reducing agent into the hemoglobin solution. The reducing agent can consume oxygen free radicals and inhibit the combination of hemoglobin and oxygen free radicals and thus inhibit the promoting oxidation of Hb.This paper utilize the special peroxidase activity of hemoglobin catalytic hydrogen peroxide oxidation tyrosine reaction to remove H2O2, and protect the hemoglobin oxygen carriers, lessen reperfusion injury.The result showed that the capacity of Hb and Tyr on consuming H2O2, and synergistic action between Tyr and Hb has antioxidant capacity and protect Hb itself. The significant protection of Tyr and Hb from H2O2-induced endothelial cell oxidative damage was observed via the oxidative damage model we established. The data of cell survival rate and LDH, MDA analyses showed that the antioxidant capacity of Hb(and Tyr) is better than Hb(or Tyr) only. As well, the result of Flow cytometry(FCM) showed that synergistic actions have the protective effect on cell damage and cell apoptosis induced by H2O2.
