Objectives: This experiment aims to1. Observ the angiogenic effect on chick chorioallantoic membrane with modified collagenmatrices by covalent incorporation of heparin and loading with Chinese herb Huangqiinjection, compound injection of red sage root, Mailuoning Injection, Xue Sai TongInjection, Shen Xiongqin injection.2. Observ synergistic effect on chick chorioallantoic membrane through implantingrhVEGF165 and Chinese herb Huangqi injection, compound injection of red sage root,Mailuoning Injection, Xue Sai Tong Injection, Shen Xiongqin injection into CAM.Methods:We Implant modified collagen matrices by covalent incorporation of heparin and loadingwith Chinese herb Huangqi injection, compound injection of red sage root, MailuoningInjection, Xue Sai Tong Injection, Shen Xiongqin injection into chick embryochoriallantoic membrane (CAM) model and observe the angiogenic effect by quantitativecounting CAM microvessels around collagen, mensurating haemoglobin by spectrophotometry, measuring dryweight of collagen, analyzing immunohistochemicalmicrotome section of collagen.Results1. No significant differences of the death rates of embryos were observed between thecontrol group of and the groups of modified collagen matrices.2. Vessels show a spoke-like structure growing towards the modified matrices(modifiedcollagen matrices by covalent incorporation of heparin and loading with rhVEGF165/Chinese herb).3. Compared with non-modified collagen matrices, the dryweight of modified collagenmatrices have significantly increased. (P, H<0.008, H+V<0.001, H+M<0.001,H+M+V<0.002)4. The microvessels counting of group of Chinese herb compound injection of red sageroot(5ul) has significantly increased, Compared with control group(P<0.001) and group ofmodified collagen matrices(P<0.008),and the counting of group of Mailuoning Injectionand Xue Sai Tong Injection has significantly increased too(P<0.001 and P<0.007,P<0.007and P<0.034). The microvessels counting of group of compound injection of red sageroot(10ul), Huangqi injection and Shen Xiongqin injection has significantly increasedcompared with control group (P<0.001 and P<0.001, P<0.008).5. The microvessels counting of group of rhVEGF165 has significantly increased,Compared with control group(P<0.004). 6. The corpuscular hemoglobin of all groups of Chinese herb has increased, greatly ingroup of Shen Xiongqin injection, compound injection of red sage root, rhVEGF 165, andXue Sai Tong Injection (P<0.001,P<0.001,P<0.001,P<0.016), compared with group ofmodified collagen matrices. The corpuscular hemoglobin of group of Mailuoning Injection,Huangqi injection, and Mailuoning addding Huangqi injection has the tendency ofrising(P<0.065,P<0.098,P<0.075).7. By analyzing immunohistochemical microtome section of collagen(CD34),we found that there are nosignificant statistical differences in number of CD34 positive cells (P>0.05)in all groups of Chineseherb,and no significant statistical differences between all groups of Chinese herb and modifiedcollagen,too. (p>0.05)Conclusions1. Modified collagen matrices has the tendency in angiogenesis,and rhVEGF165 has thegreat activity of promoting angiogenesis.2. Chinese herb Huangqi injection, compound injection of red sage root, MailuoningInjection, Xue Sai Tong Injection, Shen Xiongqin injection has great effect onangiogenesis,and this activity is similar to rhVEGF165.3. Chinese herb adding rhVEGF165 has the tendency of synergistic effect onangiogenesis.
