Effect Of Huang Pu Tong Qiao Capsule On Cognitive-behavioral And Regeneration Of Neural Cells In Rats With Cerabral Ischemia-reperfusion

Objective:To establish rats model of cerabral ischemia-reperfusion in rats and e-valuate the effects of Huang Pu Tong Qiao capsule for tonifying Qi and kidney,eliminating phlegm for resuscitation and removing blood stasis on neurobehaviora-l, learning and memory of model rats. And the effects on proliferation, different-iation, survival of neurons in dentate gyrus are also evaluated.Methods:60male Sprague-Dawley rats were randomly divided into3groups, including sham-operation group, cerebral ischemia group(blank control group) and c-erebral ischemia+Huang Pu Tong Qiao capsule group(Huang Pu Tong Qiao grou-p).Four-vessel occlusion method was used to make acute cerebral iscemia-reperfu-sion model. Each group had20rats and neurological function of each rat was gr-aded by neurological severity score. Learning and memory of rats were testedbyopen field test, locomotors active test and Morris water test.5-Bromo-2-deoxy-uri-dine injection and immunofluorescence histochemical were used to evaluate t-heeffects of Huang Pu Tong Qiao capsule on proliferation, differentiation, survivalof neurons in dentate gyrus.Results:(1) Neurological severity score: Sham-operation group had no neurological defect.Balnk control group had higher scores than sham-operation group within6h aftercerabral ischemia-reperfusion(P<0.01). Huang Pu Tong Qiao group had lower scor-es than blank control group in3rd and7th day after ischemia injury, but the di-fference is non-significantly.(2) Open field test: The outcome of7th,14th and21st day is significantly(P<0.01). Between Huang Pu Tong Qiao group and blank control group, difference ofscores in7th is non-significantly, but the14th day is significantly(P<0.05), and t-he21st day is significantly(P<0.05).(3) Hidden platform experiment: Huang Pu Tong Qiao group had obviously less duration than blank control group from3rd day to5th day after training(P<0.01).(4) Spatial probe test: Compared to sham-operation group,correct crossing freque-ncy of blank control group and Huang Pu Tong Qiao group were evidently redu-ced,the difference is significantly(P<0.05). And frequency of Huang Pu Tong Qi-ao group was more increased than blank control group(P<0.01).(5) Compared to sham-operation group, blank control group and Huang Pu TongQiao group had much more proliferation of neurons in dentate gyrus(P<0.01). An-d the difference between blank control group and Huang Pu Tong Qiao group i-s nonsignificantly(P>0.05). Both group had new cells in28th day after cerebral i-schemia, and model group had more(P<0.01).(6) In28th day after cerebral ischemia, New neurons of blank control group incr-eased in28th day after ischemia(P<0.01), and Huang Pu Tong Qiao group hadmuch more new neurons(P<0.01).Conclusions:(1) Blank control group rats had neurological impairment,and neurons were overExciting,independent activity decreased, with varying degrees of spatial learning a-nd memory ability damage;(2) Huang Pu Tong Qiao capsule can obviously improve the the spatial learningand memory,and can reduce the whole cerebral ischemia injury in rats caused byexcessive nerve excitability and improve the autonomous activity of rats;(2) Huang Pu Tong Qiao capsule can promote survival of new cell and neuronsdifferentiation.Therefore, we hypothesized that Huang Pu Tong Qiao capsule mayimprove the cognitive behavior of rats rather than promote rat hippocampal dent-ate gyrus area of new cells to survive and to the differentiation of neurons.
