Objective:To study the mechanisms and effects of Capsule for promoting memory and benefiting longevity (CPMBL) on the animal of Alzheimer' s disease, and at the same time, find out some other pharmacological and toxicological data of CPMBL. Methods:l.Make learing and memory rention model:The memory process of acquisition impaired animal model made by scopolamine intraperitoneal injection on mice.The memory process of consolidation impaired animal model made by NaN02 subcutaneous injection on mice.The memory process of retrieval impaired animal model made by alcohol. The learning and memory capacity were observed by experiments on avoiding dark going through crooked road and step down test. 2.Make AD model:A long term D-galactose subcutaneous injection on mice (120mg/kg, 6 weeks), we made learing and memory rention disorder, then water maze test, step-through test,and step down test were performed to explore the changes of the senile animals' memory capacity with the test of SOD, MDA, MAO, AchE, Ca2+, NO in the brains of the senile mice. 3 Do actue toxicological experiment of CPMBL. Results:1. CPMBL have some effects on the animal' s memory three stages, the effect of the high dose of CPMBL is the best.2. CPMBL could significantly improve the study and learning capcity of the senile mice. CPMBL can raise the brain and thymus coefficient. And the CPMBL can decrease the the level of the MDA, MAO, AchE, raise the competence of the SOD. Afer three doses of CPMBL administration, the level of the MDA dropped from 5.61 ± 1.99nmol/mgprot to 5.49 ± 1.35,4.42 ± 0.59,3.80 ± 1.46 nmol/mgprot, the level of MAO dropped from 18. 08±3. 80 U/h/mgprot to 16. 64 ±4. 67, 13. 44±4. 38,12.15±2. 04 U/h/mgprot, the level of AchE dropped from 1. 00±0. 31 U/mgprot to 0. 95±0. 43, 0. 50 + 0. 31, 0. 47 + 0. 21U/mgprot, the level of the SOD raise from 82.19 + 22.42 U/mgprot to 85.33 + 23.17,104.51 + 7. 938,132. 64 + 27. 67 U/mgprot. However, CPMBL had no obvious influence on the mice' s NO, Ca2+.It is suggested that CPMBL could reinforce intelligence by strengtheningimmunity, cleaning free radical and decreaseing AchE, raising the level of the Ach.3. Before the normal experiment, we give the most density of the CPMBL (0. 4ml/10g) to the mice, no one mice died. Because the volume and the density can not be increased, we can not measure the LD50, so we do the biggest capacity the mice can indure. The biggest capacity is 83g/kg to take orally. It is 228 times for people to take orally. Through the experiment, it has little toxicity to take orally , it is very safe for people to take orally. Conclusion:Summarizing the above-mentioned results, we can get the conlusion that the formula of CPMBL presented protective effects on various experimental models of AD, and it can decrease the level of the MDA, MAO, AchE and raise the content of the SOD in the brains of the mice obviously, Maybe the formula of CPMBL can ameliorate the memory retention disorder by acting on multiple pathways and effecing multiple targets.it is necessary for us to explore the mechanism of the formula of CPMBL. Furthermore, no acute toxic reaction was observed.It is safe to take orally.
