ObjectiveDetected the expression of hTM5and ANCA in serum and hTM5in colonmucosa of ulcerative coliti(sUC),and investigated hTM5clinical significance inUC.Materials and MethodsCollected from April2012to June2013First Clinical Hospital of ShanxiMedical University, Department of Gastroenterology outpatient and inpatient UCserum of60patients,30cases of intestinal specimens and clinical data, includingsevere UC in30patients, UC mild group,2012Guangzhou Conferencediagnosis "consensus opinion on the diagnosis and treatment of IBD norms"[1].Central peripheral blood using ELISA method myosin isoforms5antibody levelsin the control group of healthy volunteers30cases. Colonic mucosa tissues wasdetected by immunohistochemistry intestinal tropomyosin isoform5positive cellsin the control group from normal colon stump cancer underwent surgicalresection. In addition, indirect immunofluorescence assay UC60patients andhealthy controls positive rate of neutrophil cytoplasmic antibodies in30cases ofanti-serum.Result(1) immunohistochemical method: moderate to severe patients, patients withmild UC UC, colon healthy control subjects hTM5expression rate wasstatistically significant difference in epithelial cells (P <0.05).(2) enzyme-linkedimmunosorbent assay: moderate to severe patients, patients with mild UC UC,healthy control population differences hTM5in serums was statisticallysignificant (P <0.05).(3) Indirect immunofluorescence: serum ANCA-positive rate was significantly higher in patients with UC healthy control group, thedifference was statistically significant (p <0.05).(4) serum concentrations andANCA-positive rate hTM5do Spearman rank correlation analysis showed thatthe concentration of serum hTM5with ANCA-positive rate was positivelycorrelated (r=0.428.p=0.000).Conclusion(1)UC colonic mucosa of patients with UC and the expression level ofdisease severity hTM5antibodies in serum were positively correlated, suggestinghTM5affect the incidence and severity of UC.(2) ANCA combined detection of serum hTM5and contribute to thediagnosis and severity of UC judgment.
