Background and Aims:This study aimed to observe the expression levels difference of miRNA-137and miRNA-29a in lung cancer tissue and adjacent noncancerous tissue sample In non-small cell lung cancer (NSCLC) patients, explore the relationship between expression levels and clinical data,Methods:We studied frozen specimens of lung-cancer tissue from76randomly selected patients who underwent surgical resection of NSCLC. Using real-time reverse-transcriptase polymerase chain reaction (RT-PCR) analysis, we chosed U6as the internal control for real-time RT-PCR because it is invariant in clinical cancer specimens. We obtained microRNA-137and microRNA-29a expressions Jevel in76NSCLC patients and evaluated diagnoisis value and the association between the level of expression and survival.Results:We analysised76cases In90cases of NSCLC patients after surgery. After t-test (P<0.01), we obtained that miRNA-137showed high expression and miRNA-29a for the low expression in lung cancer. Then ROC curve analysis, Kaplan-Meire survival analysis and Cox regression analysis were analysised, When miRNA-137Ct (cycle threshold) value is33.34(AUC=0.753,95%CI:0.677-0.830, P<0.001), possess the best sensitivity of73.7%and specificity of65.8%and MiRNAs-29a Ct value of21.62,(AUC=0.685,95%CI:0.601-0.769, P<0.001), with the best sensitivity of48.7%and specificity82.9%. Survival analysis of miRNA-137expression and overall survival (overall survival, OS) is unrelated (P=0.775).MiRNA-29a (HR,2.704;95%CI:1.056-6.920; P=0.038) is independent prognostic factor for survival in NSCLC.Conclusions:Our microRNA signature is closely associated with diagnosis and overall survival among patients with NSCLC.
