Objective To explore a new method of establishning an animal model of obstructive sleep apnea hypopnea syndrome with hypertension(OSAHS). This animal model is very inpprtant guiding significance to study OSAHS,which is a basic disease to influence every system especially the cardiovascular system.Method40Wistar rats fed adaptively1week,and randomly divided into4groups(10in each group). A group,1ml sodium hyaluronate injected on both sides of the pharyngeal cacity and the tongue. B group, used composite high-fat lavage replication obesity model of hyperlipidemia rats. C group, method A+B. The control group only fed normal. Under light anesthesia,muzzle flow, blood oxygen saturation monitoring of sleep in A and C groups,and periodic measurement each group blood pressure at the same time,determined whether OSAHS and hypertension, measured blood liqid, angiotensin-Ⅱ (Ang-Ⅱ) and endothelin-1(ET-1) by ELISA. Dynamic observation the change of vascular active substance each group. After8weeks,HE to observe each group rats hippocampal structure.Results①under light anesthesia, A group and C group before and after building, the rats apnea index(AI), the longest apnea time and loeest blood oxygen saturation concentration between the difference had statistical significance(P<0.01),after8 weeks,C group compared with A group,3sets of indicators had statistical differences(P<0.01);②After8weeks,B and C in the two groups on high-fat diet compared with the control group,weight(g), Lee’s index and liqid levels were statistical differences(P<0.05);③Before building,each rat arterial systolic blood pressure(SBP)was not difference,and A,B,C three groups of rats SBP increased, at8weeks three groups compared with the control group and their level before building SBP had statistical significance(.P<0.05), three groups compared with each other were statistically significant;④After building, A,B,C three groups of rats serum levels of ET-1increased,at8weeks,compared with the control group,the serum levels of ET-1of three groups had statistical difference(P<0.01),and the level of C group was higher than B and C two groups(P<0.05);⑤After building, A,B,C three groups of rats serum levels of Ang-Ⅱ increased,at8weeks,compared with the control group,the serum levels of Ang-Ⅱ of three groups had statistical difference(P<0.01),and the level of C group was higher than B and C two groups(P<0.05);⑥The serum ET-1was positively correlated with SBP in A,B,C three groups,and at same to Ang-Ⅱ⑦A,B,C three groups of rats hippacampal CA3area were different degree of morphological changes.Conclusions The animal model of OSAHS with hypertension in rats established by injection of Sodium Hyaluronate is steadyand credible.
