Background and Objective:Cervical cancer is the second female cancer worldwide, and the most common pathological type of cervical carcinoma is the squamous cell carcinoma, accounting for about80%. Sustained infection of high-risk human papillomavirus is an important cause of cervical cancer, but the majority of HPV infected patients are able to clear the virus depending on the body’s own immune system, only a few patients will develop into cervical intraepithelial neoplasia, even further progression for cervical cancer. That’s means the body’s immune regulation system plays an important role in the HPV-associated tumors. A variety of immune cells, especially various types of T cells, have an significant role in the development of cervical cancer.Th17cells、Thl cells and Tc1cells are the body’s important immune cells, involved in the development and progression of many kinds of diseases including tumor. Th17and Th1cells are CD4+helper T cells, which have important influence in many diseases such as autoimmune diseases, inflammatory diseases and viral infections. Thl cells has been proved to be closely related to the development of a variety of malignancies. In recent years, researches about Th17cells and cancers have made great progress, but there are still many problems and controversy. Tcl T cells, a subgroup of CD8+cytotoxic T cell, can secrete IFN-γ and other cytokines and get involved in immune regulation. It also has important significance in tumor immune. There has been no researches about the expression and the correlation among the three kinds of T cells in cervical cancer. We aim to explore the percentages of Th17cells、Thl cells and Tcl cells in tumor tissues of squamous cell carcinoma of the uterine cervix, as well as the correlation among Th17cells、Th1cells and Tcl cells.Methods:We chose30cervical squamous cell carcinoma patients as the experimental group and17chronic cervicitis patients with benign uterine diseases as the control group. Then we collected the fresh cervical tissues when the patients undergoing hysterectomy. After being grinding and digestion, the tissues are digested into single cell suspension. We separated the lymphocytes. As we have known, T cells with CD3+CD8-IL-17+are Th17cells, and T cells with CD3+CD8-IFN-γ+are Th1cells, while T cells with CD3+CD8+IFN-γ+are Tcl cells. Then we detected the percentages of Th17cells、Th1cells and Tcl cells by fluorescence staining with flow cytometry.Result:(1) The percentage of Thl7cells was (1.51±0.46)%in the control group, while in the experimental group was (3.89±1.26)%,which was apparently higher than the control group(p<0.01).In the experimental group, the percentage of Th17cells was associated with the patients’lymph node metises(p=0.038).(2)The percentage of Thl cells was(6.86±1.82)%in the control group, while in the experimental group was (1.88±0.76)%,which was apparently lower than the control group(p<0.01). In the experimental group, the percentage of Thl cells was not associated with the patients’clinical-pathologic factors(p>0.05).(3)The percentage of Tcl cells was (5.37±2.95)%in the control group, while in the experimental group was (1.45±0.71)%,which was apparently lower than the control group(p<0.01). In the experimental group, the percentage of Tcl cells was associated with the patients’clinical stage(p=0.033)and vasoinvasion(p=0.041).(4) In the experimental group, there was a negative correlation between the percentages of Th17cells and Tcl cells (r=-0.445,p=0.014), and a positive correlation between the percentages of Th17cells and Thl cells (r=0.506,p=0.004).Whilet there was no meaningful correlation between Thl cells and Tcl cells, as well as in control group.Conclusion:Compared with the normal tissues, the proportion of Th17cells was increased in cervical squamous cell cancer tissues,while the proportions of Thl cells and Tcl cells were decreased, the proportions of Th17cells and Tcl cells might be assiociated with the patients’ prognosis. The correlations between either of the three subgroups in cervical cancer patients were different from the normal. Th17cells、Thl cells and Tc1cells played important roles in tumor immunity of cercical cancer.
