BACKGROUD Modern assisted reproductive technology for the majority of infertile couples brought the Gospel, but in the implementation of technology there are still many problems to be solved.In in vitro fertilization and embryo transplantation lab-grown stage, often meet with the following:(1) observing the oocytee fertilization, can only see a prokaryotic, known as the single pronucleus fertilization, and then develop into embryos;(2) did not meet the original nuclear, but develop into embryos. Normal fertilized ooctyee appeared two pronuclears.These two sources of embryos can be used? This has plagued embryologist, while previous studies focused on the D3day embryos. With the continuous development of assisted reproductive technology, blastocyst culture gradually mature, as a non-invasive method for screening embryos and improve pregnancy rate provides a better technical conditions. Whether can pass the extension of in vitro culture time, make the abnormal fertilized embryos to screen out good development potential for transplantation?OBJECTIVE To explore the value of blastocysts derived from gametes with different numbers of pronucleus by compare their blastulation rate and clinical pregnancy rate.METHOD Blastulation rate, good-quality blastocyst rate and clinical pregnancy rate in CET cycles of blastocysts derived from2PN,1PN or OPN gametes were compared in IVF/ICSI-ET cycles. Will source OPN blastula biopsy of trophoblast cells fixed FISH after testing.RESULTS In IVF cycles, blastulation rate of embryos from1PN gametes were lower that of embryos from2PN or OPN gametes(P<0.01),no significant difference between2PN and OPN(p>0.05). In ICSI cycles, blastulation rate of embryos from1PN or OPN gametes were lower that of embryos from2PN gametes. There was no significant difference in clinical pregnancy rate between the three kinds of blastocyst. FISH detection31blastocyst; Each of these three films were blastocysts signal for18cases, both for15cases of diploid chromosome detection, ZhengChangLv83.3%,3cases of abnormal detection, the abnormal rate was (16.7%). More than13cases for part of fixed, fixed part is normal in11cases, ZhengChangLv was84.6%, the fixed part anomaly in2cases, the abnormal rate was15.4%.CONCLUSION Blastocysts derived from1PN/OPN gametes and blastocysts from2PN gametes could be treated equally. They could be transferred back to patients on the basis of prenatal diagnosis.
