Objective: To investigate the relationship between single nucleotidepolymorphisms(SNPs) of PRKCG gene(rs3745406, rs2547362) andosteosarcoma susceptibility in the osteosarcoma patients and the normalpopulation..Methods: Sixty-one patients with osteosarcoma who had been admitted inour hospital from January2011to December and63healthy adults were enrolledin this study. A2-ml peripheral blood sample was taken form each participant.The RT-qPCR method was used to detect the genotype and allele frequencydistribution of PRKCG gene at rs3745406and rs2547362in osteosarcomapatients and normal population.. Osteosarcoma patients were divided into servalgroups according to the clinical parameters such as age, gender, histology, tumorlocation, Enneking classification, tumor metastasis and therapy, and then weanalyzed the relations between the genetic polymorphism and clinical parameters.Results:(1) The results shown that the genotypes of PRKCG gene atrs3745406included CC, CT and TT. The differences of genotypes(CC, CT, TT)and the alleles(C, T) frequency distribution at rs3745406site were notstatistically insignificant between the osteosarcoma patients and normalpopulation.(P=0.490, P=0.554).(2)The genotypes of PRKCG gene atrs2547362included CC, CT and TT. The differences of genotypes(CC, CT, TT)and the alleles(C, T) frequency distribution at rs2547362were statisticallyinsignificant between the osteosarcoma patients and normal population.(P=0.006,P=0.007).(3) The differences of genotypes(CC, CT, TT) and the alleles(C, T)frequency distribution at rs2547362were statistically insignificant betweenpatients with metastasis and patients without metastasis(P=0.000, P=0.000). TheCT and TT genotypes and the T allele carrier frequency at rs3745406werehigher in patients with metastasis than in patients without metastasis. SNPs atrs2547362were not associated with clinical parameters.Conclusions: The genetic polymorphism of PRKCG gene at rs2547362areassociated with osteosarcoma susceptibility. The TT genotype and T allele atrs3745406are associated with metastasis of osteosarcoma,which may be a riskfactor for metastasis in the osteosarcoma patients.
