Background and objectives: Nasopharyngeal carcinoma (NPC) ismalignant tumors in southern China with one of the most common thehighest incidence and mortality rate in the world,and it shelvesrepresent18%of all malignant tumors,rangking eighth in allmalignant.It poses one of the most serious public health problemsin this area. Radiotherapy is the primary treatment forNPC.However,the radio-resistance of nasopharyngeal carcinoma cellsis one of the main factors that restrict its curative effect andprognosis. The molecular mechanism underlying radioresistance of NPChas not been elucidated yet. The finding of radioresistant proteinswill be useful for revealing the mechanism of NPC radioresistance,and providing the scientific basis for predicting NPC sensitivity,which plays an important role in improving efficacy and prognosisof NPC.This study use the pre-induced radio-resistance of NPC cells To establish a GFP-tagged radioresistant xenograft tumor model ofhuman nasopharyngeal carcinoma (NPC), and build the foundation forfurther study.Methods: By means of lentivirus to establish a stable GFP-expressedRadioresistant human NPC cell lines: GFP-CNE-2R cell lines andGFP-expressed non-radioresistant human NPC cell lines: GFP-CNE-2cell lines.Its radiosensitivity was measured by using a clonogenicsurvival assay. In order to establish xenograft tumor the cell lineswith stable expression of GFP were subcutaneously implanted intoBALB/c-nu nude mice. The tumors-bearing mice received X-rayradiation when the tumors reached1cm in size,10Gy in1fraction.In vivo imaging technology was used to take images of tumorproliferation and metastasis.Results: The stable GFP-expressed GFP-CNE-2R cells and GFP-CNE-2cellline were successfully established, the clone formation assaysshowed GFP-CNE-2R cell lines were successfully givenradioresistance, All the xenograft tumor models of GFP-CNE-2R cellsand GFP-CNE-2cells were established successfully, and the tumorcells possessed the morphological characteristics as human NPCcells,Compared to the GFP-CNE-2tumors,the GFP-CNE-2R tumors growth was not inhibited by irradiation.Conclusion: The CNE-2R xenograft tumors are much radioresistant thanthe CNE-2xenograft tumors.
