Objective To investigate the correlation of fluorescence intensity ofEGFP and expression of insulin.Methods Construct recombinant plasmid prIns1EGFP and transfectHIT-T15cells, the cells were cultured and randomly divided into three groups,low glucose group(LG)、 middle glucose group(MG) and high glucosegroup(HG),The released insulin and A single islet beta cell relative fluorescenceintensity at baseline and after4hours stimulation were measured bychemiluminescence analysis(CA) and fluorescence microscope methodrespectively, protein level was detected with immunocytochemistry.Results The level of insulin and a single islet beta cell relativefluorescence intensity on the HG groups was higher than LG group(P<0.05),theexpression level of proinsulin on the HG group was higher than LG group (P<0.05),after the glucose stimulation,, there was significant change of asingle islet beta cell relative fluorescence intensity in HG group(P<0.05),withina certain range of glucose concentration, compared fluorescence intensity andprotein level at the corresponding time shows that they have positive correlation.(r=0.896)(P<0.05).Conclusions The fluorescence intensity of EGFP and level of insulinhave good correlation.
