This thesis comprises two parts: Part I: Expression of S100A8and S100A9protein in human plasma and its relation with NPC clinical stages and with EBV-VCA-IgA Level; Part II: The expression of S100A8and S100A9in tissues ofnasopharyngeal carcinoma.PART1: Expression of S100A8and S100A9Protein in Human Plasma and ItsRelation with NPC Clinical Stages and with EBV-VCA-IgA LevelObjective To explore the correlation of the expression levels of S100A8andS100A9protein in the plasma of patients with nasopharyngeal carcinoma (NPC)with the clinical stages and the Epstein-Barr virus (EBV) VCA-IgA antibody.Methods ELISA method was used to examine the concentrations of S100A8and S100A9and the levels of EBV-VCA-IgA in the plasma of patients with NPC(stage II:36cases; stage III:104cases; stage IV:104cases) and healthy people (104cases). The results were analyzed by SPSS16.0statistical software.Results The concentrations of S100A8and S100A9in the plasma of NPCpatients were0.44(0.25,0.78) ng/mL and202.71(185.79,238.54) pg/mL, whichwere significantly higher than those of healthy people [0.29(0.18,0.44) ng/mL and 192.43(170.66,193.42) pg/mL](P<0.05).The each concentration of S100A8andS100A9in the plasma for stages II, III and IV of NPC patients were all higher thanthose of healthy people (P<0.05),and those of stage II and III were higher thanthat stage IV (P<0.05); the concentration of S100A9at stage III was higher thanthat of stage II (P<0.05). Correlation analysis showed the concentrations ofS100A8and S100A9in the plasma of NPC patients had no correlation with thelevel of EBV-VCA-IgA (P>0.05), but there was positive correlation betweenS100A8and S100A9, P <0.05.Conclusion The expressions of S100A8and S100A9protein in the plasma ofNPC patients were upregulated,and associated with the clinical stages of NPC.PART2: The expressions and clinical siginificane of S100A8and S100A9intissues of nasopharyngeal carcinomaObjective: To investigate the relationship between the expressions of S100A8and S100A9in tissues of nasopharyngeal carcinoma(NPC), to lay a foundation forthe further functional study.Methods:(1)Immunohistochemical SP staining was used to detect theexpression of S100A8and S100A9in24cases of non-keratinizin(undifferentiated)NPC with diagnosed preliminarily (stage Ⅱ:5cases, stage III:9cases, stage IV:10cases) and6cases of nasopharyngeal mucositis. The association of clinical datawith S100A8and S100A9expression was examined. The results were analyzed bySPSS16.0statistical software.(2)Western blot was used to detect the expression ofS100A8and S100A9in3cases of non-keratinizin(undifferentiated) NPC withdiagnosed preliminarily and1case of nasopharyngeal mucositis (16.67%and50%).The each positive expression rate of S100A8and S100A9in the tissues for stages II, III and IV of NPC patients were all higher than nasopharyngeal mucositispatients.Results: The immunohistochemical results showed S100A8and S100A9protein mainly distributed in the cytoplasm; Fisher’s exact test showed The totalpositive rate of S100A8and S100A9in the tissues of NPC patients were70.83%and91.67%respectively, which were higher than those of nasopharyngealmucositis(16.67%and50.00%)(P<0.05). Statistic analysis found significantdifference between the stage Ⅲ of NPC and nasopharyngeal mucositis tissues.There were no significant difference between the stage Ⅱ, Ⅳ of NPC andnasopharyngeal mucositis tissues.(2) Western Blot experiments showed,theexpression levels of S100A8and S100A9in biopsy of nasopharyngeal carcinomawere higher than those of nasopharyngeal mucositis tissues.Conclusion: Upregulated expression of S100A8and S100A9may beassociated with the development and progression of NPC.
