ObjectiveTo investigate the effect of Yulangsan polysaccharides (YLSPS) on immunefunction,Anti-stress ability,related indicators of blood in immunosuppressivemice induced by cyclophosphamide.To preliminary study it’s mechanism further,and provides experimental basis for the research and application of YLSPS.Methods:1Effect of Yulangsan Polysaccharides on the immune function ofimmunosuppressive mice induced by cyclophosphamide60SPFKMmice,20g±2g,were divided into six groups randomly:the controlgroupcyclophosphamide (CTX)model group,Levamisole (LMS) group,and high,medium,low doses of Yulangsan polysaccharides (YLSPS)groups.The micewereadministated with drug for10days continuously,Except the control group,othergroups were constructed by hypodermic injection with CTX (30mg/kg) everyother day to establish immunosuppressive mouse model.At the last day ofgavage,the mice were fasted for12h, and then were executed,the thymus andspleen were weighed,after that its thymus index and spleen index were calculated.The activities of Lactate dehydrogenase (LDH) and acid phosphatase(ACP) in the spleen were assayed too.The phagocytosis ability of macrophage inliver and spleen were measured aswell.DTH induced by DNFB was alsoobserved too.2Effect of Yulangsan Polysaccharides on Anti-stress in immunosuppressivemice induced by cyclophosphamide Experimental animal’s handling methodwas taken as above.After60minutes at the lastadministration,the impact onanti-stress action by mice’s swimming test,anti anoxia test,lower temperatureresistant test and higher temperatureresistant test,nitrite poisoning anoxia testwere observed.The anti-stress ability in immunosuppressive mice were detected.3Effect of Yulangsan Polysaccharides on the index of blood inimmunosuppressive mice induced by cyclophosphamide Experimentalanimal’s handling method was taken as above.After the last medication,themice were fasted for12h,then were executed by cervical dislocation,the bloodwas taken from the mice’s eyeball,the index of RBC,Hb,WBCand PLT in theblood of mice were measured.The content of blood serum’s IgA,IgM,IgG weredetected by using Immunoturbidimetry.Results:1Effect of Yulangsan Polysaccharides on the immune function ofimmunosuppressive mice induced by cyclophosphamide Compared with theNS group,indexs of thymus and spleen of the CTX group were reducedsignificantly after administration(P<0.01),Compared with the CTX group,indexsof thymus and spleen in all doses of YLSPS groups werepromoted(P<0.05).Compared with the NS group,the activities of Lactatedehydrogenase (LDH) and acid phosphatase (ACP) of the CTX group werereduced significantly in mice spleen after administration(P<0.01),the activities of LDH and ACP in the medium and high doses of YLSPS groups were elevatedobviously in immunosuppressive groups(P<0.01).Compared with the NSgroup,the phagocytosis ability of macrophage in liver and spleen of the CTXgroup were reduced significantly after administration(P<0.01),the phagocytosisability of macrophage in liver and spleen in the medium and high-doses ofYLSPSgroups were elevated obviously in immunosuppressive groups(P<0.01).Compared with the NS group,Ear swellings of the CTX group were reducedsignificantly after administration(P<0.01),Ear swellings in the medium and highdoses of YLSPS groups were elevated obviously in immunosuppressivegroups(P<0.01).2Effect of Yulangsan Polysaccharides on Anti-stress inimmunosuppressive mice induced by cyclophosphamide Compared with theCTX group,the swimming time and the resistant of lower temperature’s time inall doses of YLSPS groups were promoted(P<0.05),especially in the high dosesof YLSPS groups(P<0.01).At the same time,resistant of higher temperature’stime in the medium and high doses of YLSPS groups wereelevated(P<0.05).Compared with the CTX group,the anti-anoxia in all doses ofYLSPS groups were promoted(P<0.05),especially in the high doses of YLSPSgroups(P<0.01).And the nitrite poisoning anoxia ability in the medium andhighdoses of YLSPS groups were improved(P<0.05).3Effect of YulangsanPolysaccharides on the index of blood in immunosuppressive mice induced bycyclophosphamide Compared with the CTX group,the numbers of RBC wereincreased in the medium and high doses of YLSPS groups that caused by CTX(P<0.05),the level of Hb was improved in the medium and high doses of YLSPSgroups,especially in the high doses of YLSPS groups(P<0.01).The number ofWBC was improved in all doses of YLSPS groups that caused byCTX,especially in the medium and the high doses of YLSPS groups(P<0.01).And the level of PLT in the high doses of YLSPS groups wasimprovedtoo(P<0.05).Compared with the CTX group,the content of IgA in boththe medium and the high doses of YLSPS groups were improved(P<0.05).Thenumber of IgG in all doses of YLSPS groups were increased(P<0.05).Meanwhile,the number of IgM in all doses of YLSPS groups were increasedtoo(P<0.05),especially in the high doses of YLSPSgroups(P<0.01).Conclusions:1Effect of Yulangsan Polysaccharides on the immune function ofimmunosuppressive mice induced by cyclophosphamide YLSPS couldstrengthen non-specific and specific immunologic functions ofimmunosuppressive mice induced by CTX.2Effect of YulangsanPolysaccharides on Anti-stress in immunosuppressive mice induced bycyclophosphamide YLSPS could increase the action of anti-stress obviously.3Effect of Yulangsan Polysaccharides on the index of blood inimmunosuppressive mice induced by cyclophosphamide YLSPS couldimproved the ability of Cell-mediated immunity and humoral immunity inimmunosuppressive mice that induced by CTX.
