ObjectClarify the role of SIDT2in gastric cancer. Provide some novel ideas and rationales for the study and treatment of gastric cancer. which can be used for the diagnosis and prognosis of gastric cancer.MethodsThe level of SIDT2were been detected in gastric cancer cell lines SGC-7901,BGC-823,MKN-45,MGC-803and normal gastric mucosa epithelial cell GES-1by Real-time PCR and Western Blot. After using si-SIDT2to knockdown the SIDT2expression in SGC-7901, we detected the prolifertion and apoptosis by MTT and FCM respectively.ResultsThe SIDT2mRNA expression of MGC-803and MKN-45is higher than that of GES-1. On the contrary, the SIDT2mRNA expression of SGC-7901and BGC-823is.lower than that of GES-1. The SIDT2protein of GES-1did not detected, but that of diverse gastric cancer cells increased in varying degrees. si-SIDT2did not effect on proliferation of SGC-7901cells by MTT assay, and did not participate in the apoptosis process of SGC-7901cells by FCM.Conclusions1、SIDT2may relate with the occurrence and development of gastric cancer, and may be an oncogenesis gene.2、SIDT2showed a negative correlation in differentiation gastric cancer cells.3、SIDT2may have no effect on proliferation and apoptosis of SGC-7901cell.
