Objective:1. To evaluate the clinical efficacy of Compound Xinyi and to confirm its therapeuticmechanism.2. To detect the role of CD34+and IL-5in the allergic rhinitis and the influence of drugtherapy on this pathway.3. Finally, this study provides convenient and quick indicators to evaluate the clinicaltherapeutic effect.Methods:From February to December2013,60patients with allergic rhinitis were collectedfrom the First Affiliated Hospital of Anhui Medical University. In addition, a normalgroup was set, among which10healthy people from medical center.60patientsrandomly grouped into two groups depending on compound xinyi nasal or azelastinehydrochloride nasal spray. In the former group, compound xinyi nasal drops were usedthree times a day, each time on each side of the nasal cavity of a drop (equivalent to30uL); in the later group, patients were treated with azelastine hydrochloride nasal spraytwo times per day, each time on each side of the nasal cavity of a spray (equivalent to0.56mg). After four weeks treatment, The content of IL-5in serum of peripheral bloodand bone marrow was measured by ELISA,and count the numbers of CD34+inhematopoietic progenitor cells by flow cytometry. Signs of nasal mucosa of patientswere tested by immunohistochemistry and clinical manifestations were observed toevaluate the therapeutic effect of compound xinyi nasal drops.Results:1. Treatment effctsAfter four weeks of treatment, among the30patients of treatment group, the total effective ratio of83.3%, as for the control group, the total effective ratio of86.7%.Compared two groups, there has no statistical significance(p>0.05).2. The changes of symptoms and signsBefore treatment,the difference of symptoms and signs between two groups has nostatistical significance (P>0.05). On the contrary, after treatment, the symptom indexeswere reduced significantly with statistical significance(P<0.05).3. The numbers of CD34+hematopoietic progenitor cells and IL-5in serum.Before treatment, the numbers ofCD34+hematopoietic progenitor cells and IL-5inserum of60patients were significantly higher than the healty group (P<0.05). Aftertreatment, the numbers of CD34+hematopoietic progenitor cells and IL-5in serumwere reduced significantly with statistical significance compare with the healthy group(P<0.05).4. CD34+cells of nasal tested by IHCCD34+cells mainly express on the submucosa, the number of CD34+cells weresignificantly higher after treatment than before(P<0.05).Conclusion1. It is obvious of compound xinyi nasal drops to ameliorate the symptoms of the2. This test suggested that the level of CD34+and IL-5play an important role inallergic rhinitis. The level of CD34+hematopoietic progenitor cells and IL-5inserum could be reduced by the compound xinyi nasal drops through negativefeedback pathway of marrow.3. The test provided a simple and valid method to evaluate the clinical therapeuticeffect.
