Objective:To extract the peripheral blood and marrow from the Japanese White Rabbit, to in vitro separate, purify and culture CD34+Cells, and to investigate their biological characteristics.Methods:By the Percoll gradient centrifugation and adherence screening method, the CD34+Cells were separated from the blood and marrow, we counted the cultured cells and analyzed the cell growth curve; We numbered blood smear group, albuginea of blood smear group, marrow smear group, and albuginea of marrow smear group. After the immunohistochemical staining, we investigated the CD34+Cells and counted the density of cells. Then compared the difference of each group.Results:From the seventh day on, cultured the blood and marrow, characterized by exponential phase of growth with strong proliferation, growth curve showed typical "S" shape; The CD34+Cells from marrow grow better than from blood; After the immunohistochemical staining, resulted the positive expression was distributed in the cell membrane; The CD34+Cells from blood was the colour of straw, the CD34+Cells from marrow was the colour of dark brown; Counted the density of cells, compared the difference of each group, we could find out(Top-Down):albuginea of marrow smear group contained the most the CD34+Cells, the next is albuginea of blood smear group and marrow smear group, blood smear group contained the least.Conclusion:(1) The result suggest the CD34+Cells are feasible to be used in the correlated experimental study from the seventh day on.(2) The number of the CD34+Cells from marrow are higher than from blood. The CD34+Cells from marrow grow better than from blood(3) The immunohistochemical staining showed:the blood and marrow from the Japanese White Rabbit contained the CD34+Cells, resulted the positive expression was distributed in the cell membrane. The expression of marrow'CD34+Cell showed strong positive, the expression of blood'CD34+Cell showed Weak positive.(4) By the Percoll gradient centrifiigation and adherence screening method,, we can separate more CD34+Cells.
