Objective: To study the normal rabbit and rheumatoid arthritis fibroblast-like synovialcell proliferation differences; research hypericin’s Photodynamic effect to kill rabbitrheumatoid fibroblast-like synovial cells.Methods: To establish ovalbumin-induced arthritis rabbit model, and to take themodel and normal rabbit knee joint synovial cell culture, using MTT assay to surveythe growth curve of the two cell population, to curve fitting the growing data use SPSS18.0software, calculate doubling time of the two cell population,then compare thedoubling time difference between them; selection of complete medium and incubatedwith rabbit class hypericin concentration gradient of rheumatic synovialfibroblast-like cells, with hematoporphyrin monomethyl ether positive control, thenPDT, using MTT assay to survey the relative number of surviving cells in eachgroup,to analysis by SPSS18.0software significant differences between the groupsafter each set of data collation, and make a significant difference reasonablenessanalysis concluded.Results: Rabbit rheumatoid synovial fibroblast-like synovial cells doubling time areshorter about5hours than normal cells, normal rabbit knee joint synovial cellpopulation doubling time is34.3hours, and FLS cell doubling time is29.5hours;hypericin’s complete medium concentration in0.78mg/L, illumination time of90minutes, the rabbit knee arthritis fibroblast-like synovial cells have a more stronginhibition and its concentration between1.56to50mg/L, illumination time of90minutes, can kill the FLS.Conclusion: Rabbit rheumatoid synovial fibroblast-like synovial cells were strongerproliferation and rapider proliferation than normal cells in vitro proliferative capacity;hypericin can inhibit or kill rabbit knee joint rheumatoid arthritis fibroblast-like synovial cells at a certain time and lighting conditions, hypericin’s photodynamiceffect are relat to the lighting time and hypericin’s concentration closely.It has a gooddevelopment prospects that to treatment rheumatoid arthritis.
