Objective:To investigate the impact and possible mechanisms of fenofibrate on THP1-derived foam cells cholesterol content.Methods:THP1established by culturing cells and THP1-derived foam cells, experiments were randomly divided into four groups:control group, foam cell+25umol/L fenofibrate, foam cell+50umol/L fenofibrate, foam cells+100umol/L fenofibrate; using oil red O staining, identification THP1-derived foam cells; detection of adenosine triphosphate by RT-PCR-binding cassette transporter A1(ABCA1), ATP-binding cassette transporter G1(ABCG1) mRNA expression, with Western blotting assay cells in each group ABCA1and ABCG1protein content.Results:After THP1-derived foam cells by oil red O staining, seen in red cell lipid droplets; compared with the foam cell groups, different concentrations of fenofibrate group (25umol/L,50umol/L,100umol/L) of ABCA1, ABCG1mRNA and protein expression was significantly increased, the difference was statistically significant (P <0.05), and with the increased concentration of fenofibrate, ABCA1expression, ABCG1protein in a dose-dependent; compared with the foam cell groups, different concentration of fenofibrate group (25umol/L,50umol/L,100umol/L) cholesterol levels, significantly reduced, the difference was statistically significant (P<0.05), and with fenofibrate concentration increased cholesterol levels showing a dose-dependent.Conclusions:Fenofibrate may stimulate the expression of ABCAl, ABCG1mRNA and protein to reduce cholesterol levels in the foam cells, promote cholesterol efflux.
